The Pathogenesis of Stagnated heat Competing syndrome,which is Propose by Professor Zhou Zhongying, has great Significance in the Differential Treatment.The research of Stagnated Heat Competing syndrome and its relationship with systemic lupus erythematosus, is divided into two parts, theoretical aspect and experimental study. Theoretical part:to further explore the pathogenesis of blood stasis and Stagnant Heat, we used literature consult, since it is the basic pathogenesis of systemic lupus erythematosus to discuss the feature,definite Stagnated heat Competing syndrome, and further explore Stagnant Heat. Method:Ds-DNA, ANUA, S-P2 was determined with enzyme linked immunosorbent assay(ELISA); MMP-9, TNF-a was determined with immunohistochemistry assay, gene expression of MMP9, TNF and TNF-a was determined with RT-PCR in MRL/lpr lupus mice treated by cooling blood Stasis,24-hour urine proteinuria was also analysized. Results:The ds-DNA, ANUA levels of MRL/lpr mice model group were significantly higher than those of the negative group, the proteinuria level was slso higher than those of the contrasting group. MMP-9, TNF-a in kidney of model group,and the gene expression of MMP-9, TNF-a, VEGF of model group was higher than those of negative group。After the treatment of Western medicine (CTX, prednisone), traditional Chinese medicine (cooling blood side)and both the Chinese and Western medicine, proteinuria in the treated group was significantly less than those in control group.However, the proteinuria reduction of the Western medicine group is superior to the Chinese medicine group (P<0.05), and that of the combination of Chinese and Western medicine group is superior to the Western medicine group and Chinese medicine group (P<0.05).The decrease of MMP-9, TNF-αof the combination of Chinese and Western medicine group is superior to the Chinese or Western medicine group. Both chinese andWestern medicine can reduce the gene expression of MMP-9.The effect of the combination of Chinese and Western medicine group and the Western group is superior to traditional Chinese medicine group. Drug treatment can interfere with TNF gene transcription and the effect of the combination of Chinese and Western medicine is more evident than that of Chinese or Western medicine. Drug treatment can also interfere with VEGF gene transcription and the effect of western medicine is more evident than both the Chinese medicine and combination of Chinese and Western medicine. The research implied that cooling blood and removing blood stasis treat systemic lupus erythematosus patients by reducing mmp-9, TNF-αin the kidney, decreaing mmp-9, TNF-α, VEGF gene expression as part of the mechanism of action. To drug testing card, the traditional Chinese medicine can reduce ds-DNA, TNF-αin the blood;ds-DNA, TNF-αcan serve as a molecular biomarker of heat stasis.
