To observe the hearing protection of herbal compound for reinforcing kidney,activating blood and arousing consciousness(HCRAA) in gentamicin(GM)-induced ototoxic mice, analysising the expression of Neurotrophic Factor 3(NT-3) of the Cochlear Nucleus,Inferior Colliculus and Auditory Cortex of central auditory system Iter in damaged internal ear and approaching correlated mechanism of multi-target action of traditional Chinese medicine.Herbal compound for reinforcing kidney,activating blood and arousing Consciousness (co-mposition:Drynariae,epimedium,astragalus,Chinese angelica,Salvia,Chuanxiong,Acorus grami-neus,leech, magnets,etc.) were decocted for high,medium and low concentrations of the three decoction containing respectively pharmacognostic 2.375g/ml,0.475g/ml,0.095g/ml (based on the mouse body weight by 25g,0.8ml of intragastric administration every day, the decoction of the concentration of one is equivalent to 70kg adult day clinical routine the equivalent amount of volume, high concentration and low concentration of decoction were respectively equivalent to the amount of 5 times and 0.2 times the dose).40 Kunming mice were randomly divided into 5 groups of 12. A, B, C, D group:be respectively taken HCRAA high, medium and low concentration and normal saline by of every day, 100mg/kg, for 15 consecutive days. From the first day of GM intraperitoneal injection, four groups be respectively intragastric administratered for high, medium and low concentration of Chinese compound decoction and normal saline, for 20 days; E groups:contrast group, normal feeding, not any treatment. After administration the mice in each group were measured auditory brainstem evoked potentials (ABR) thresholds, and adopted the method of immunohistochemistry to analysis the expression of Neurotrophic Factor 3(NT-3) of in auditory center by computer image analysis system.Expression of NT-3 in the Cochlear Nuclecus and Inferior Colliculus of mice:HCRAA of high and middle concentration could elevate the expression of NT-3, compared in these two groups and with normal contrast group, the result had no statistics significance (p>0.05); However, the expression of NT-3 of normal saline group and HCRAA of low concentration were lower than normal contrast group, the result had no statistics significance (p>0.05) in these two groups, compared with normal contrast group and those two groups of HCRAA of high and middle concentration, the result had statistics significance (p<0.01). Expression of NT-3 in the auditory cortex of mice:the expression of NT-3 in all the groups were general, the result had no statistics significance (p>0.05).HCRAA could effectually antagonize ototoxicity of GM and protect the auditory center. HCRAA could elevate the expression of NT-3 in auditory center, obviously improve hearing and play an important action in protection the integrity of hearing iter when taking ototoxicity medicine meanwhile taking HCRAA medicine.
