This paper is supported by committee of pharmacopeia of China, No.YD-070. We make a draft of quality standards of Aspongopus.; provide some solutions to solve adulterate problems; carry out studies on pharmacodynatics according to the clinical effect of Aspongopus.; to study Aspongopus.'s chemical characteristics, we use IR, PAGE and SDS-PAGE. The main research contents and results are as follows:1. We observe two kinds of insect:Aspongopus. and its imitation Cyclopelta perva Distana and make a comparation of their character properties and microscopic charateristies. We find a new difference in their character properties which is an important character to make a distinction between the two bugs. Their microscopic charateristies are distinguished primarily by one kind of setal alveolus which Aspongopus. does not has but its imitation Cyclopelta perva Distana has. The discovery are effective for quality control of Aspongopus. and its Chinese medicines. The result can complement and perfect the microscopic charateristies of Aspongopus. in "chinese pharmacopoeia".2. We analyses chemical quality of Aspongopus. Thin layer chromatography is used to identification fatty acid of Aspongopus. We make corelation analysis between total ash, total moisture and dilute ethanol extracts. The result shows that total ash and total moisture is meaningful to control the quality of Aspongopus.3. We get an adulteration component from one of poor quality batches of crude drugs, it is magnesium sulfate. By using coordination titration, their contents were determined. We describe character properties of these poor quality batches and quick chemical identification is established.4. The infrared spectrum was used to identity Aspongopus. During the scope of 4000-500 cm-1, the petroleum ether extract of Aspongopus. have seven common peaks and their degrease powder have nine common peaks. By using IR, we can identify Aspongopus. adulterate with magnesium sulfate or foreign oil.5. The content of protein in Aspongopus. is determined by BCA method. There is big difference in batches and the content of protein in crude drugs is affected by freshness, adulteration and storage. Thinking from the edible and a nutritional point of view, batches 1 (Guizhou),2 (Guizhou),5 (Yunnan) and 12 (Zhaotong,Yunnan) are in good quality, the content of protein is from 7.33% to 8.50%.6. Systematic solvent method is used to extract Aspongopus. The water solution and different fraction from the ethanol extracts of Aspongopus. are used to test anticoagulant activity. We study the clotting time by using glass-tube method in vivo test and experiment the platelet aggregation of rabbit's blood which induced by adenosine diphosphate in vitro test.The different position extracts from Aspongopus. all have anticoagulant activity. So the anticoagulant activity is the joint action of water-soluble part and the liposoluble part of Aspongopus.The water solution of Aspongopus. can raise CT of normal and blood stasis mice and restrain rabbit platelet aggregation.7. Electrophoresis (PAGE and PAGE-SDS) are used to identify protein of Aspongopus. The protein electrophoresis bands of Aspongopus. are distinct and the method can be reproducible but these two methods can not distinguish Aspongopus. from its imitation Cyclopelta perva Distana at present.This paper provide some simple and practical solutions and have some reference values for the research of the quality identification and the quality control of Aspongopus. The results laid the groundwork for establish the method of evaluating the quality of Aspongopus. and provide scientific experiment evidence to ensure the safety of clinical use.
