| ObjectiveBcr-abl negative (MPN) has always been a difficult clinical treatment. In 2005, the discovery of the mutation of JAK2 V617F helped to get a new understanding of the pathogenesis of bcr-abl negative MPN. The clear definition of the molecular mechanism accelerated the research process of the MPN on molecular targeted inhibition therapy. Many kinds of JAK2 inhibitors are under a variety of pre-clinical studies, and some have entered clinical trials.The preliminary results have shown that there exists inhibition of the signaling pathways related to JAK2 both in vivo and in vitro.However, the R & D costs and treatment costs are relatively high, and its predictable side effects can not be ignored.Currently, the study on the anti-tumor effect of traditional Chinese medicine has been a hot issue in clinical research in our country. Though combined chemotherapy remains the main anti-tumor therapy, it has inevitably more side-effects. Moreover, there is primary or secondary drug resistance in the process of treatment. It is necessary to seek new anti-tumor drugs which have higher therapeutic effect and milder side-effect. Traditional Chinese medicine has been accepted by more and more experts and patients and is effective in the combined therapy of malignant tumor. Clinical experiments have shown that, the traditional Chinese medicine can not only decrease the toxicity and increase antitumor efficiency of radiotherapy and chemotherapy, but also prevent the development of precancerous lesion and has anti-tumor effect. It has many advantages of mild side effects, abundant resources, cheap price and long history application. There is a long history by using traditional Chinese medicine to cure tumor in our country. The treatments we use are different from Western Medicine and have the great advantages in anti-tumor. Therefore, research on the theorem of traditional Chinese medicine's anti-tumor is of great importance in illustrating the basic theory of traditional Chinese medicine, clinical therapies and the researches of traditional Chinese medicine. Marsdenia tenacissima(Roxb.)Wight et Arn, which is bitter, cool and a little sweet, mainly grows in Yunnan and Guizhou province. It was first recorded in the "Materia Medicapolyanthum".It can stop coughing and gasping, eliminate inflammation, relieve pain and has the effect of lactogenesis and diuresis.It was used to treat chronic bronchitis. It is concerned by a great deal of medical workers that using Marsdenia tenacissima to treat cancer can achieve positive effect. The objective of this test is to study and prove that traditional Chinese medicine Marsdenia tenacissima's anti-tumor mechanism and investigate the mechanism of anti-tumor from the facts of apoptosis.2.MethodsMTT deoxidized method was applied to survey the inhibition effect of Xiaoaiping on human leukemia HEL bearing JAK2 mutation, in vitro. HEL is co-cultured with different concentration of Xiaoaiping extracts.Flow cytometry was applied to detect the ratio of Annexinâ…¤positive cells and to reflect the apoptotic rate induced by drugs.SPSS 11.0 software was used to carry out statistical analysis. T-test and x2-test were employed. The size of test was p<0.05.3. ResultsHigh concentrations of Xiaoaiping injection (60,70,80,90ul/ml respectively) has been evaluated for their inhibitory effects on HEL cells. And the anti-proliferative effect was dose-dependent. It showed significant difference through statistics analyzing (P<0.01).High concentration (50,75ul/ml)of Xiaoaiping injection has no significantly accelerated effect on the apoptosis of HEL cells. And continue to addition of concentration(100ul/ml) can only promote cell death directly.4. ConclusionHigh concentration of Xiaoaiping injection has determinate inhibitory effect on HEL cells. And the effection is dose-dependent.Xiaoaiping injection has no determinate effect on the apoptosis of HEL cells...
