Objective:To investigate the role of quercetin (Que) in the proliferation and the expression of actin-a (a-SMA) of transforming growth factor-β1 (TGF-β1)-induced neonatal rat cardiac fibroblasts (CBF) in vitro.Method:Neonatal rat CFBs were isolated and cultured in vitro.The influence of quercetin on cardiac fibroblast proliferation was determined by WST-1 assay.The cardiac fibroblast cell cycle was analysised by flow cytometry. The expression of a-SMA was detected by immunocytochemistry (SABC).The experiment was divided into three parts. PartⅠ:To investigate the effect of QUE on the proliferation of cultured rat CFBs in basic state, there were four groups,blank control group and different concentration (50μmol/L,25μmol/L,12.5μmol/L). PartⅡ:In order to observate the effects of Que on the TGF-β1-induced cardiac fibroblast proliferation and expression of a-SMA, the experiments were divided into five groups,blank control group, TGF-β1(5μg/L) group and different concentration (50μmol/L,25μmol/L,12.5μmol/L)+TGF-β1(5μg/L) groups. PartⅢ:In this part,there were four groups to investigate the influence of quercetin on cardiac fibroblast cell cycle, TGF-β1 (5μg/L) group and different concentration(50μmol/L,25μmol/L,12.5μmol/L) +TGF-β1(5μg/L) groups. SPSS 13.0 package used.OD values of cell activity are showed as the mean and standard deviation (x±s),the single-factor analysis of variance for statistical analysis,chi-square test, were compared with the standard four-cell table method to examine the level of the P<0.05 was statistically significant. Results:(1)QUE has no significant effect on the cardiac fibroblast cells proliferation in the basic state; (2)QUE could inhibit the cardiac fibroblast cells growth stimulated by TGF-β1 in a concentration-dependent manner; (3) QUE could inhibit the expression of a-SMA of TGF-β1-induced cardiac fibroblast cells in a concentration-dependent manner. (4) QUE could prevent TGF-β1-induced cardiac fibroblast cells from G0/G1 phase to S phase in a concentration-dependent manner,QUE various concentration of cells in S phase was significantly lower than TGF-β1 group (P<0.05);Conclusion:1. QUE can not inhibit CFBs proliferation in basic state,but can inhibit CFBs proliferation induced by TGF-β1 in a concentration-dependent manner;2.QUE could prevent TGF-β1-induced cardiac fibroblast cells from G0/G1 phase to S phase and inhibit its expression of a-SMA in a concentration-dependent manner.This may be one of the ways of QUE inhibiting myocardial fibrosis induced by TGF-β1.
