Effects Of Sorafenib Cmbine With Doxazosin To Hormone-refractory Prostate Cancer DU145 Cells In Vitro

Objective: To study the Effects of sorafenib cmbine with doxazosin to hormone-refractory prostate cancer DU145 Cells In Vitro.Methods: The DU145 cells were divided to 4 groups as control group,sorafenib group,curcumin group and sorafenib-doxazosin group. sorafenib group and curcumin group were divided to 5 gradients of concentration(3.125,6.25,12.5,25,50μmol/L), were dealed with different time (24h,48h,72h). by MTT assay at each time after the addition of the different drugs to DU145 culture and the OD values were computed, to calculate the inhibition ratio. Different concentration of sorafenib cmbining with Different concentration of doxazosin were deal with different time (24h,48h,72h), to measure the OD values , to calculate the inhibition ratio and q values .Spectrophotometry measure DNA content, microscope observe the change of morphologic of cell, Immunocytochemistry measure for influence of caspase3 and PCNA .All the results (the inhibition ratio,Cell Cycle, the expression of protein of proliferation and apoptosis)were analysed by SPSS 13.0 .Results: Sorafenib cmbine with doxazos have collaboration to hormone-refractory prostate cancer DU145 Cells...
