Objective:To study DNA ploidy and expression of HER2 and FOXM1 in patients with gastric carcinoma and to explore the mechanism of invasion and metastasis of gastric carcinoma.Methods: We systematically analysed the expression of HER2 mRNA and FOXM1 mRNA in human gastric carcinomas (n = 20) and normal tissues (n = 20) by reverse transcriptase PCR. Additionally, we investigated HER2 and FOXM1 expression in human gastric carcinomas (n = 88) and normal gastric tissues (n = 20) by immunohistochemistry (IHC).Finally, we analysed DNA ploidy in human gastric carcinomas (n = 88) by Flow cytometry(FCM). SPSS version 13.0 was used for statistical analysis.Results: HER2 and FOXM1 were found to be overexpressed in gastric cancinomas in comparison to normal gastric tissue both on the RNA and protein level(P<0.01). Positive correlation between HER2 and FOXM1 both on the RNA and protein level was observed(P<0.01).Expression of HER2 and FOXM1 protein level was not correlated with gender,age and the degree of differentiation(P>0.05),but was correlated with lymphatic metastasis and the depth of invasion (P<0.05).And the rate of DNA heteroploid was not correlated with gender and age (P>0.05),but was correlated with the degree of differentiation,lympathtic metastasis and the depth of invasion (p<0.01).Position correlation between the expression of HER2 and FOXM1 on protein level and the rate of DNA heteroploid was observed too(P<0.01). Conclusions:1.The abnormal expression of HER2 and FOXM1, DNA aneuploid are correlated with lymphatic metastasis and depth of invasion of gastric carcinoma.2. Positive correlation is found between HER2 and FOXM1。And positive correlation is found between the expression of HER2,FOXM1 and the rate of DNA heteroploid too.
