Hyperlipidemia is one of the diseases in modern civilization society. With the quality improvement of life,and more opportunity to eat high protein and high fat,coupled with reduced physical activity, the blood fat cannot be consumed induce to the Hyperlipidemia. In China ,hyperlipidemia population increase along with economic development, and the younger trend be worried. Medical profession has proved—Hyperlipidemia patients who suffer from heart disease is four times than the normal human. According to statistics ,China now has 9,000 people with hyperlipidemia at least. A large number of research data show, Hyperlipidemia is an independent and important risk factor to stroke, coronary heart disease, myocardial infarction. In addition, Hyperlipidemia is also an important risk factors of promoting hypertension, glucose tolerance and diabetes. Hyperlipidemia can also cause other serious diseases, such as fatty liver and so on.Traditional Chinese medicine in the treatment of hyperlipidemia compared to Western medicine has its unique advantages, it has positive effects, continuous, and little side effects.Particularly in terms of differentiation, reflect the flexibility of Chinese medicine prescriptions. There is no the hyperlipidemia name in Chinese traditional medicine. In treatment on hyperlipidemia Chinese traditional medicine usually use the herbal tonifying the liver and kidney, strengthening the spleen and replenishing qi, and removing heat to loosen bowels.Hedan tables is a Chinese complex prescription developed by famous Chinese doctor Yang Jisheng. It has identify effect in treating with hyperlipidemia. This compound is composed by lotus leaves, Salvia miltiorrhiza, hawthorn, senna and Psoraleae. Lotus leaves is the king drug can qi dampness; Salvia as minister drugs, improve blood circulation, Salvia as minister drugs, strengthen the liver and kidney; hawthorn as adjuvant drug, promoting digestion; senna as messenger drug, reducing heat stagnancy. This medicine have significant clinical effects.In recent years, the research on Hedan tables is not very much, a small number of reports focused on their hypolipidemic effect and clinical observation. There were rarely reports on its composition and hypolipidemic active ingredients. As a scientific and objective analysis and quality control, identifying the components of Hedan tables, researching the relevant of these components'metabolic changes in vivo with hypolipidemic effects, is significance in finding efficacy components and its quality control. The study of pharmaceutical components in Chinese medicinal herbs demands not only the extract quantitative analysis of target compounds, but also the characterization of the compounds, and eventually control and evaluate the quality of Chinese medicinal herbs. It demands not only the differentiation of chemical compounds, but also the characterization of the compounds in plasma, and the possible pharmaceutical components were detected. It demands not only the differentiation of active compounds in plasma, but also metabolic and action mechanism of the compounds in plasma, and the possible pharmaceutical components were detected, which evaluate the pharmaceutical action and select as pharmaceutical components.It was necessary for pharmaceutical analysis to use multiple LC-MS techniques, because that the single analytical technique could not fulfill the demand of Chinese medicinal herbal analysis for its complexity and entirety. In this research, based on modern analytical techniques, from the Chinese medicinal herbs of Hedan tables ,we developed several method and strategies of characterization of chemical components in Chinese medicinal herbs and preliminary screening of pharmaceutical active components, established the fingerprints of chemical components in vivo and in vitro, and apply these method and related analytical techniques in whole quality control of Chinese medicinal herbs. There were 41 components in Hedan tables identified based on the characterization and structure elucidation, and unambiguous fingerprints in vitro were developed.17 components come from lotus leaves, 10 components from salviae miltiorrhizae, 8 components from psoralea, 5 components from hawthorn and 1 component from senna. This results agree with traditional theory.Hedan tables composed five herbals, and a large number components in it, even many isomer. In order to differentiate these isomer, we adjusted the fragment volt of TOF/MS. Deduced the composition according to the fragment ions at different volts. In the end we can not only know the molecular formula through the accurate molecular weight, but also differentiate these isomers at TOF/MS.During the components research in Hedan tables, we find that there are many alkaloids in lotus leaves ,many salvia quinones in salviae miltiorrhizae, many coumarins in psoralea. According to the decomposition rule of the component, we find that RDA cleavage is the main manner of alkaloids, and many Salvia quinonoids can lose CO. Base on the identification of chemical components in Hedan tables, oral administration Hedan tables to SD rats, using HPLC-TOF/MS, identify the chemical components in plasma of rats, and we find 11components in it.A quantitative method of four components in Hedan tables, including nuciferine, tansinone IIA, psoralen and isopsoralen were developed by HPLC-DAD, which reflect contents in Hedan tables, control and evaluate the quality of Chinese medicine. The results indicated that this method can be used to quantify the Hedan tables which provide a basis for advancing the quality control standard of Hedan tables.
