| Clinically, there is a wide range of narcotics used for dogs, but each has its own advantages and shortcomings. Inhalant anesthetics has stable effect and high depth of anesthesia, but due to its high price and complex use, it's seldom used clinically. Thus, the research and application of combined anesthesia have been concerned, but these combined anesthesia can not meet the complex requirements of animals,. Therefore, ideal combined anesthesia for dog should be studied as soon as possible. In this study, based on the theory of animal anesthesia and physiological characteristics of dogs, the combined anesthesiawere studied reasonablly and systematically.In pre-test, xylidinothiazoline,qiangtongning,midazolam,droperidol were selected to be the component of the combined anesthesia; According to the results of four factors-three level orthogonal tests and decisive orthogonal test with mice, the optimal ratio for the component is 2.5:0.2:0.4:0.25 (V:V), prescription is tentatively named xylidinothiazoline combined anesthesia;Toxicity test were carried out on mice to investigate the safety. The results were ED50=7.41 mg/kg,LD50=64.57 mg/kg and AI= LD50/ ED50=8.71, indicating that the combined anesthesia has great margin for safety;Breathing monitor, fetal monitor and EEG monitor were used to investigate the effects of combined anesthesia on dogs, the results showed that the indicators were in the normal physiological range;The operation test showed that xylidinothiazoline combined anesthesia had good effects of sedation, analgesia and muscle relaxation; Compared with QMB anesthesia in dogs, the results showd that xylidinothiazoline combined anesthesia had less effects on circulation, breath and brain function than QMB, which indicated that xylidinothiazoline combined anesthesia had advantages over QMB.In summary, xylidinothiazoline combined anesthesia has good effects of sedation, analgesia and muscle relaxation, less effects on physiological function, wide safety range, and can be used in clinically. |
