Objective To observe the expressions and changes of IL-15,IL-17 in the synovial fluid of knee after trauma in rabbits and the Cartilage and synovium degenerated.This article is aimed to design their characteristics in order to define the destructive process of post-traumatic osteoarthritis.Methods Totally 48 healthy rabbits were divided into experimental group and control group randomly with 24 in each group. The experimental groups was cut off the anterior, posterior cruciate ligament, medial collateral ligament, and full medial meniscus of the left- posterior knee, another 24 rabbits received unilateral arthrotomy as pseudo operative controls. The specimens were collected on 6 hour, 12h ,24h and week 1 ,week 2 ,week 3 ,week 4 after surgery respectively. the enzyme - linked immunosorbent assay (ELISA) was performed to measure the expression of cytokines IL– 15,IL– 17 .The morphological change of cartilage and synovium were investigatedResults The expression of IL -15, IL -17 was seen with peak values on 6 hour compared with other time points after operation(P<0.05).the association between time and concentration is inverse proportion,compared with the 24 hour. The expression of IL -15, IL -17 on week 1 was decreased Obviously(P<0.0). The expression of IL -15, IL -17 was increased from 1 week to 4 week. The expression of IL -15, IL -17in experimental group was significantly higher than those in control group(P<0.05)Articular cartilage lost original luminosity,The surface of articular cartilage exhibited gray-yellow .HE staining showed Surface layer not smooth ,small crevasse, hyaline degeneration ,fiber tissue hyperplasia ,some cartilage matrix metachromatic was weaken , the toll number of chondrocyte decreased , the partial number of chondrocyte increased cartilage matrix fibrosis. synoviocyte hyperplasiaConclusions At the early stage of post- traumatic osteoarthritis, the expressions of interleukin-15,interleukin-17was upgraded. the concentration changes to follow time and time Cartilage and synovium was degenerated , interleukin-15,interleukin-17 is closely involved in the early stage of post- traumatic osteoarthritis...
