Objective: To observe the expression changes of cytokine SUMO-1 in the formation of hepatic fibrosis in rats , and investigate the role of SUMO-1 in hepatic fibrosis.Methods: Ninety male SD rats were randomly divided into two groups: model group and control group. The rats in the model group were given 40% CCl4 by subcutaneous injection (0.3ml/ 100g), the rats in control group were given equivalent volume of physiological saline. The liver tissue was taken out for experiment at 2 nd,4 th,6 th,8 th,10 th and 12 th week respectively , hepatic fibrosis were detected by pathology method. SUMO-1mRNAs were detected by RT-PCR and SUMO-1 proteins were detected by Western blotting, the data of each group were analyzed and compared with each other, and compared with the control group.Results: With the formation of hepatic fibrosis, the expressions of mRNA in normal(0.721±0.006),2nd(0.856±0.007),4th(1.092±0.085),6th(1.273±0.010),8th(1.565±0.012),10th(1.782±0.014),12th(2.025±0.016)week were gradually and significantly increased, and the expressions protein of SUMO-1 in normal(0.758±0.060),2nd(0.922±0.073),4th(1.170±0.093),6th(1.370±0.109),8th(1.613±0.128),10th(1.823±0.145),12th(2.078±0.165)were gradually and significantly increased, compared with the same time control group. ( P < 0. 001)Conclusions: the expressions of mRNA,and protein of SUMO-1 were gradually and significantly increased,it may play an important role in the formation of hepatic fibrosis.
