Objective The aim of the present study was to examine an association of brain- derived neurotrophic factor(BDNF) polymorphisms and the response to modified electroconvulsive therapy (MECT) in patients with major depressive disorder (MDD).Method The patient group were recruited from the mental health center of the first affiliated hospital of Chongqing medical university. All 110 patients met the diagnostic criteria of DSM-IV for major depression, meanwhile they consecutively admitted for eight MECT in the Department of Psychiatry. The control group 100 people from visitors of the medical center, interns and internship students of the first affiliated hospital of Chongqing medical university respectively. About 5 mL of blood are drawn from a vein in the subjects' inner elbow region,then EZ-10Spin Column Genomic DNA Isolation Kit,polymerase chain reaction (PCR) and gene sequencing were used ultimately identify the genotype between the patient group and the control group by Statistical Analysis. The Hamilton depression scale (HAMD) was used as an outcome measure of depression after eight MECT. The BDNF gene rs6265, rs7103411 genotype and allele frequency were counted after MECT between the responders and the non-responders.Results Neither of these BDNF polymorphisms have significant differences between the healthy control group and major depressive disorder patients. And the two Single nucleotide polymorphism sites are not associated with treatment of response to MECT. In addition, no significant differences were found in the genotypes concerning the MECT response and allele of rs7103411, however, significant differences in the A allele frequency of rs6265 were found(χ~2=5.250,Р=0.022,OR=1.740,95%CI: 1.022~2.963). A allele of rs6265 site have better responded to MECT.Conclusion No correlation between major depression disorder and polymorphism of BDNF gene rs6265, rs7103411 was found in the whole population. rs6265 site A allele of BDNF gene is relevant with the therapeutic efficacy of MECT in patients with major depressive disorder.
