Objective To investigate the effects of procyanidins on proliferation and apoptosis of cisplatin-resistant human overian cancer cell line COC1/DDP and to explore the molecular mechanism .Methods (1) COC1/DDP cells were cultured in the presence of procyanidin (at 0,20, 40,80,160,320ug/ml )After 24, 48, 72h of culture ,the morphological changes of COC1/DDP were examined, and the inbition of cell proliferation was masured by MTT assay .(2) Cell apoptosis was masured by flow cytometry. Reverse transcription-polymerase chain reaction (RT-PCR)and Westrn-blot were performed to detect the expression of bcl-2,bax and caspase-3.Results Observe the COC1/DDP cells incubated in presence of GSPE by light microscopy, HE staining,MTT. GSPE inhibited the growth of COC1/DDP cells in vito in a concentration and time-dependent manner. Flow cytometry analysis showed that procyanidins induced apoptosis of COC1/DDP cells and the percentage of apoptotic cells increased with the concentration of procyanidins administered and also the elongation of treatment time.The expressionof bax and caspase-3 were upregulated and associated with a downregulation of the expression of bcl-2 in the course of COC1/DDP apoptosis when treated with GSPE.Conclusion The proliferation inhibition and proapoptosis activity of GSPE,which is significantly observed in the human overian cancer cell lines COC1/DDP in vitro,may be related to the increased caspase-3 expression and the rate of bax/bcl-2.
