Objective To investigate the interaction between DKK1 gene andβ-catenin, illuminate the molecular mechanism in Wnt signaling pathway and the effect of DKK1 gene in malignant menaloma cell line A375.Methods We design three groups in our research: Ad-DKK1-GFP transfected group,Ad-GFP transfected group and blank group.RT-PCR and immunohistochemisty Assay methods were used to detect the expression of DKK1 andβ-catenin in malignant menaloma cell line A375 which transfected by adenoviruses with DKK1 gene. Cell Count,MTT and principle of statistics on quality were used to investigate the interaction between DKK1 and the malignant menaloma cell line(A375)Results RT-PCR and Immunohistochemisty analysis: The expression of DKK1 was upregulated andβ-catenin was deregulated after transfected Ad-DKK1-GFP.Cell Count: Ad-DKK1-GFP transfected group is lower than other groups.MTT: Ad-DKK1-GFP transfected group is lower than other groups. Principle of statistics on quality: Ad-DKK1-GFP transfected group is the smallest of all.Conclusion 1. Malignant menaloma cell line A375 can be transfected by adenoviruses with high expression of DKK1 gene and protein. 2. DKK1 can inhibitβ-catenin. 3. DKK1 can inhibit melanoma cells increasing.
