Objective: To explore the relation between endothelial lipase (EL) and vascular remodeling of the main artery and its possible mechanism and outcomes of atorvastatin intervention by detection the dynamic changes of EL ,nuclear transcription factor-κB (NF-κB)and vascular remodeling of the main artery in two-kidney one-clip (2K1C) hypertensive rats.Methods: Renal hypertensive rat models were constructed through two-kidney-one-clip(2K1C) technique.Thirty-eight rats were randomized into sham operation group (Sham, n=7), 2K1C 4W group (n =7), 2K1C8W group(n=7), 2K1C 12W group(n=7), atorvastatin-treated group(ATV, n=7). Atorvastatin treatment(30mg/kg/d) was started four weeks after model construction and lasted 8 consecutive weeks, given through lavage. All rats were evaluated caudal systolic blood pressure(SBP),the level of serum lipids, microscopic changes in vascular structure and ratio of intima media thickness to lumen diameter(MT/LD) of thoracic aorta. Expression of EL and nuclear transcription factor-κB (NF-κB ) were assessed with immunohistochemical staining, expression of EL-mRNA with Real-time quantitative RT-PCR.Results: High-density lipoprotein cholesterol(HDL-C) levels of 2K1C 8W and 2K1C 12W groups was significantly lower than Sham group;MT/LD of 2K1C groups was significantly higher than the Sham group, and was gradually increased with time extended;the expression of EL, EL-mRNA, and NF-κB in the 2K1C groups was significantly higher than the Sham group, and was gradually increased with time extended;the expression of EL, NF-κB were positively related with MT/LD, negatively with HDL-C. As compared with 2K1C 12W group, MT/LD and the expression of EL,EL- mRNA and NF-κB of ATV group were significantly decreased, HDL-C level significantly increased.Conclusions: (1) The significantly increased EL expression of the main artery are closely related with vascular remodeling, and may be one of the mechanisms of vascular remodeling. (2) Atorvastatin was shown to inhibit the development of vascular remodeling in renal hypertensive rats, possibly associated with decreased expression of EL.
