PARTⅠBIOLOGICAL EFFECTS OF LIPOPOLYSACCHARIDE, TRANSFORMING GROWTH FACTOR-Β1 ON MURINE BONE MARROW-DERIVED DENDRITIC CELLSObjective: To explore method of stimulating murine bone marrow-derived dendritic cells by lipopolysaccharide(LPS), transforming growth factor-β1( TGF-β1)and to study their biological character.Method: Murine bone marrow-derived dendritic cells were cultivated with cytokine GM-CSF and IL-4 for 6 days. BMDC was stimulated by medium (Control group), LPS (LPS group), TGF-β1 (TGF-β1group),LPS+TGF-β1(LPS+TGF-β1group),for 48 hours respectively.Morphological characters of BMDC were observed by a inversed microscope.surface molecules such as CD11C,CD80,CD86 and MHCⅡwere detected by flowcytometry.Interleukin-6 and interleukin-12p70 in co-culture medium was quantified by ELISA.Result: In LPS group DC presented the most typical DC morphology with the highest expression of CD80,CD86 and MHCⅡ, and they were 98.71%,95.26%,99.02% respertively.and the strongest ability in mixed lymphocyte reaction.and higher level of IL-6 and IL-12p70 compared with control,TGF-β1,LPS + TGF-β1(P<0.05).While in TGF-β1 group DC presented the less typical DC morphology with the lower expression of CD80,CD86 and MHCⅡ,and they were 46.84%,42.16%,62.52% respertively.and weaker ability in mixed lymphocyte reaction.and lower levels of IL-6 and IL-12p70 compared with control and LPS (P<0.05).Conclusion: LPS can stimulate maturation of BMDC in its late differentiation which makes it presents a more significant biological characteristics.TGF-β1 can inhibit maturation but not differentiation of BMDC and thereby can prevent its biological characteristic presentation. PARTⅡDIFFERENT EXPRESSION OF TGF-ΒR1 ON MURINEBONE MARROW-DERIVED DENDRITICCELLS, BETWEEN MICES WHICH HAVE DIFFERENT GENETIC BACKGROUND OF ASTHMAAbjective: To explore murine bone marrow-derived dendritic cells expression difference of TGF-βR1 between Balb/C mice and C3H/HeJ mice,which have the different genetic background of asthma.Method: BMDC were cultivated with cytokine GM-CSF and IL-4.Morphological characters of BMDC were observed by a inversed microscope.surface molecules such as CD11C,CD80,CD86and MHCⅡwere detected by flowcytometry. then extract the total RNA of DC,and then whether express differences of TGF-βRⅠbetween the two kinds of DC is existed or not were detected by Real time Q-PCR.Result: BMDC of Balb/C mice and C3H/HeJ mice presented the most typical DC morphology.the expression of CD11C,CD80,CD86 and MHCⅡby flowcytometry,Balb/c mice were 75.09%, 91.60%, 82.98%, 91.54% respertively.and C3H/HeJ mice were 74.73%, 92.88%, 85.39%, 91.93% respertively.so these can guarantee the purity and maturity of BMDC. and the total RNA can guarantee the purity and concentration. express differences of TGF-βRⅠbetween the two kinds of DC is existed were detected by Real time Q-PCR .Conclusion: There are different expression of TGF-βRⅠbetween BMDC of Balb/C mice and C3H/HeJ mice.
