A Research Of Expressing Of MHC-â… ,Foxp3 In Iim Patients's Musclelature

IIM is a set of acquired and autoimmunity concerned somatic muscles disease,whose diagnostic symbol is CD8,MHC-â… . CD4+CD25+Treg is one of subgroup of T cell,whose mean function is to adjust the activation and increment of the self-T cell.It does an irreplaceble action on maintaining autoimmune tolerance and preventing autoimmune disease.The expression of Foxp3,mainly in Treg cells,is thought to be the major factors determining the immune regulative function of Treg.ObjectivesTo discover the value and possible pathologic mechanism of MHC-I and Foxp3 in the diagnosis, and explore the pertinency between their express and the serious extent of the patients, through observing the distribution and the expression of the MHC-I in the IIM patients's musclelature and the distribution and the expression of Foxp3 in the inflammatory cells.MethodsThe change of their pathologic structure is observed through muscle biopsy, which is carried out on 15 IIM patients chose from those who are according with the clinical diagnosis. Comparing with the non-inflammatory myopathy patients, the distribution and the expression of the MHC-I in the IIM patients's musclelature and the distribution and the expression of Foxp3 in the inflammatory cells are observed by using immunohistochemical method.Results1. MHC-I expresses definitely in the IIM patients's musclelature, the positive rate of the 15 IIM patients in this experiment is 100%.2. Although there are not evident infiltration of inflammatory cells in 6 of the 15 IIM patients's muscle fiber, muscle enzyme abnormity and clinical symptom is still showed up in them.3. The extent of MHC-I's express in the IIM patients's musclelature and the patient's creatase are positively correlated, which has the statistical significance.4. There are 5 cases among the 15 IIM patients, whose Foxp3 in muscle fiber expresses positive. The blood serum muscle enzyme is lower in the patients whose Foxp3 express positive than those negative, and the extent of MHC-I's express is weaker and the clinical symptom is less severe.Conclusion1. The molecule of MHC-I class can be used as one of the index in the IIM's early diagnosis.2. The express of the MHC-I in the IIM canbe earlier than the inflammatory cell infiltrate.3. The pathogenetic condition of the patients is correlative with the Foxp3's express: the positive-express'pathogenetic condition are less severe, while the nagtive-express'pathogenetic condition are more serious.4. The number or disfunction of the Foxp3Treg cells might be one of the reason, which cause the IIM.
