The Expression Of Ubiquitin Proteasome System And P27 Protein In Hepatocellular Carcinoma And Its Clinicopathologic Significance

Background:Ubiquitin protein (Ub) is a highly conserved protein in eukaryotic cell.The ubiquitin protease system is an important pathway for the intracellular proteins degradation.The polyubiquitinated proteins can be degraded by UPS.The polyubiquitin is an basic accommodation mode for cell to keep dynamic balance.UPS is essential for many cellular processes.UPS can degrade intercellular proteins with high effect and selectivity,especially for cellular physiology and pathology,including cell multiplication and apoptosis, gene transcription, signal transduction, angtigen presentation, DNA damage and repair.In recent surveys,as an inhibitor of cyclin dependent kinase,p27 protein plays an important role in cell differentiation and cell multiplication.It is very closely relationship of UPS and p27 protein degradation,within Rectal Cancer, Gallbladder Cancer ,Breast Cancer,et al.But the regulative mechanism of UPS to p27 protein degradation is not great comprehension. We use immunohistochemical and western blotting method to detect the expression of ubiquitin and p27 protein in hepatocellular careinoma and its clinical significance with different clinicopathologic degrees. Then we try to find how Hepatocellular careinoma occurrence,development and prognosis.Objective:To study the relationship of the expression of ubiquitin proteasome system and p27 protein in Hepatocellular careinoma and its clinical significance with different clinicopathologic degrees.Then we try to find how Hepatocellular careinoma occurrence,development and prognosis.Methods:1. We use immunohistochemical method to qualitation detect the expression of UPS and p27 protein in hepatocellular carcinoma with different clinicopathologic degrees.2. We use western blotting method to semiquantitative detect the expression of UPS and p27 protein in hepatocellular carcinoma with different clinicopathologic degrees.3. We use Nuclei-Cytoplasm Extraction method to get nuclei and cytoplasm proteins in hepatocellular carcinoma with different clinicopathologic degrees,and then to detect the expression of UPS and p27 protein.Results:1. In immunohistochemical method detection,ubiquitin and p27 protein were expressed in hepatocellular carcinoma. Ubiquitin protein was located in nuclei and/or cytoplasm.Ubiquitin protein expression was negative correlation with different clinicopathologic degrees in hepatocellular carcinoma (p<0.05).P27 protein was mainly located in nuclei.P27 protein expression was positive correlation with different clinicopathologic degrees in hepatocellular carcinoma (p<0.05). Ubiquitin and p27 protein expression were significantly correlated with the clinicopathologic degrees(p<0.05).2. In western blotting method detection, ubiquitin protein expression was negative correlation with different clinicopathologic degrees in hepatocellular carcinoma (p<0.05). P27 protein expression was lower in poorly differentiated HCC tissue than in well-differentiated and moderately differentiated HCC tissue (p<0.05).3. In Nuclei-Cytoplasm Extraction method detection, ubiquitin and p27 protein were expressed lower in hepatocellular carcinoma cytoplasm than in total hepatocellular carcinoma cell.But in hepatocellular carcinoma nuclei,ubiquitin protein expression was negative correlation with different clinicopathologic degrees in hepatocellular carcinoma (p<0.05). P27 protein expression was positive correlation with different clinicopathologic degrees in hepatocellular carcinoma (p<0.05).Conclusion:In hepatocellular careinoma,ubiquitin protein expression was negative correlation with different clinicopathologic degrees. But p27 protein expression was positive correlation with different clinicopathologic degrees.We can find that the relationship of expression of UPS and p27 protein is very closely. Then we try to find how Hepatocellular careinoma occurrence,development and prognosis.
