Objective To explore the the effects of Chinese herbal recipe Huoxue huayu decoction (HXHYD) on BDNF,HO-1,GLAST protein expression in acute ocular hypertension of ratsMethods 90 healthy and clean Wistar rats were averagely devided into 2 groups at random, including group A and B.Group A was used for hematoxylin and eosin stain(HE stain). Group B was used for Western Blotting. Each Group was devided into 9 small groups, and all left eyes were taken. Take group A for example, A0 was normal control group, A1-A4 were intraocular hypertension groups, A5-A8 were intraocular hypertension involved HXHYD groups. Groups A1-A8 were operated acute hypertension model surgery. Then, groups A1-A4 didn't received any dispose, were sacrificed with intraperitoneal injection of excess chloral hydrate and were taken left eyeballs at 1,3,7and 14 days after the surgery. Groups A5-A8 were administered HXHYD by gavage (2ml, twice time per day) immediately after the surgery, sacrificed and taken left eyeballs at 1,3,7and 14 days. The disposes of group B were similar. All rats of group B were taken retinas of their left eyes.Results 1,Light microscope:After 1 days of acute intraocular hypertension, full-thickness of retia were dropsical severely, the nucleus of RGCs were swollen; 3d after pressurizing, the dropsical condition of retinal was improved than that of 1d, the number of RGCs was decreasing; 7d after pressurizing, the dropsical condition was disappeared generally, the number of RGCs continued to descrease, the thickness of retia was thinning; 14d after pressurizing, all constructions of the retina were disordered, the retinal became atrophy. The changes of retinal shape and the degree of the injury in the pressurizing and pouring HXHYD groups were less severe than the pressurizing groups during the same time stage.2,Western Blotting:BDNF:The expression of BDNF protein in group B1 was increased rapidly compared with normal group and got to the peak(P<0.01), descended in group B3(P<0.01), dropped to the lowest lever in group B14 which was lower than normal control group. The expression of BDNF protein in the pressurizing and pouring HXHYD group were more than those in the pressurizing groups with the same time stage(P<0.01).HO-1:In the pressurizing group, the expression of HO-1 protein was upregulated and peaked 1d after the surgery(P<0.01). After 3 days, the level of HO-1 protein declined to the normal. Then, it increased again and more than normal level on day 7(vs control, p<0.01). Still higher than normal level even at 14d. The change of HO-1 protein expression in the pressurizing and pouring HXHYD group had the same trend, but the expressions were more with the same time stage.GLAST:The expression of GLAST protein in group B1 increased rapidly (vs control, P<0.01), declined in group B3 and still higher than normal level (P<0.01). In group B7, the expression declined to the normal level which was maintained in group B14. In groupB1', B3', the expressions were higher than B1,B3 separately.Conclusion The Huoxuehuayu decoction can protect the rat's retina which was injured by acute intraocular hypertention by means of promotting the expression of BDNF,HO-1,GLAST proteins in retina.
