Deguelin is one of the most potent rotenoids. It can kill insects and delay development. In recent years, researchers have found its new function that it has antineoplasmic activity.It is reported that deguelin has chemoprevention effect to skin cancer, breast cancer, colon cancer and lung cancer. As for as now, it is unclear that the mechanism of deguelin's antitumor ability and delaying embryo development.This paper is focusing on studying the antiangiogenic effect of deguelin and the mechanism of delaying embryo development. The content is listed below.The second chapter is focusing on the antiangiogenic effect of deguelin. When treated with 0.5μM deguelin from 12hpf, the zebrafish ISV was missing. Using 0.1mM,0.2mM,0.4mM deguelin treated chick embryos and counted the number of vessel around carrier. The result was 4.50±0.79,2.30±0.67,0.75±0.71 while control was 14.54±0.93 and then count P by T-test(P<0.01). The data show very significant differences between the deguelin-treated group and the control group. According to the result, deguelin has antiangiogenic effect.The third chapter is focusing on deguelin delaying zebrafish embryo development. 0.15μM,0.8μM deguelin can delay zebrafish embryo development since 4.3hpf and 0.8μM deguelin treated embryo stay 4.3hpf. Zebrafish embryos start epiboly at 4.3hpf. So deguelin can arrest embryo epiboly. We conjecture the reseason is deguelin affecting zebrafish embryo cell migration.The fourth chapter is focusing on cell migration. After treating by 0.4μM deguelin, the number of cell permeating transwell was 28.7±6.1 while control was 121.0±4.6 and then count P by T-test (P<0.01). The data had very different. According the result, deguelin affected HUVEC migration.The fifth chapter is focusing on the mechanism of degeulin delaying embryo development. grk2 mRNA, cyp11a1 mRNA, jak1 mRNA, pou5f1 mRNA were tested by in situ hybridization and the expression level of grk2 mRNA iwas significant decreased. Then we tested grk2 mRNA by Real Time PCR, grk2 mRNA treated by deguelin was 0.013 fold of the control. Microinject grk2 mRNA into zebrafish embryo to rescue zebrafish developmet. To sum up, this paper prove that deguelin has antiangiogenesis effect through zebrafish model and CAM; degeulin can affect the migration of HUVEC. deguelin can decrease grk2 mRNA through in situ hybridization and real time PCR.
