Objectives:To observe the content of BACE1 and Aβand the effect of DNLA on BACE1 and AP in hippocampal neurons of rat with chronic hypoxic and hypoglycemic culture in vitro.Methods:The hippocampal neurons was isolated and cultured and identified in vitro. At 8th day, the cell was randomly divided into normal control groups which was cultured in normal condition, solventia control groups which was added with DMSO(0.01ng/ml) and cultured in normal condition, DNLA control groups which was added with high dosage of DNLA and cultured in normal condition, hypoxic and hypoglycemic groups which was cultured in hypoxic and hypoglycemic condition,and DNLA treatment groups which was treated with DNLA and cultured in hypoxic and hypoglycemic condition. Activition of hippocampal neurons was measured by means of MTT in vitro.The content of BACE1 and Aβwas respectively detected with radioimmunoassay and ELISA.Results:Compared with normal control group,the activition of hippocampal neurons in solventia control group was not significantly difference(P>0.05),but the activition of hippocampal neurons in hypoxic and hypoglycemic groups was significantly decreased (P<0.01).The activition of DNLA treatment groups was significantly increased than that of in hypoxic and hypoglycemic groups (P<0.01).Compared with normal control group,the content of BACE1 and Aβwas significantly increased in hypoxic and hypoglycemic groups (P<0.01). In hypoxic and hypoglycemic groups,the content of BACE1 and Aβwas significantly increased in 36h group than that of in 12h group.Compared with hypoxic and hypoglycemic groups,the content of BACE1 and AP was significantly decreased in DNLA treatment groups (P<0.01) Conclusion:The content of BACE1 and Aβwas significantly increased and DNLA can decrese the content of BACE1 and Aβin hippocampal neurons with hypoxic and hypoglycemic culture in vitro...
