Objective:To make the models of hormone refractory prostate cancer (HRPC). To assess laws of HGMB1(high mobility group protein B1,HMGB1) expression after cryoablation, and to assess HGMB1 on the impact of the anti-tumor immune response.Materials and Methods:Mouse models of HRPC were made by subcutaneouly injection of RM-1 prostate cancer cells, and the tumor-bearing mice were randomly divided into different groups:control group, cryoablation group, EP intervention treatment group. Cryoablation was applied by cryosurgery needle with 1.7mm diameter from Endocare Argon-Helium cryosurgery system. EP were given intraperitoneal injection 3ml at 12h,24h,48h respectively after cryosurgery. The number and maturation changes of dendritic cells in tumor, spleens were assessed by flow cytometry,which were sacrificed before and after cryosurgery1,7,14and21 days respectively. The number of dendritic cells surface TOLL-4 receptor were assessed by flow cytometry,which were sacrificed before and after cryosurgeryl,7,14and21days respectively.Result:(1) Cryoablation group, the levels of HMGB1 in tumor, spleens were gradually increased after AHCS,7days reached the highest value,but 21 days returned to preoperative levels (P<0.05). EP intervention treatment group, the levels of HMGB1 in tumor, spleens were gradually increased after AHCS,7days reached the highest value,but 21days returned to preoperative levels (P<0.05). control group, the levels of HMGB1 in tumor, spleens were steady (P>0.05); (2) Cryoablation group, The number and maturation changes of dendritic cells in tumor, spleens were were gradually increased after AHCS,7days reached the highest value,but 21 days returned to preoperative levels (P<0.05). EP intervention treatment group, The number of dendritic cells surface TOLL-4 receptor in tumor, spleens were gradually increased after AHCS,7days reached the highest value,but 21days returned to preoperative levels (P<0.05) control group, The number of dendritic cells surface TOLL-4 receptor in tumor, spleens were steady (P>0.05);(4) Relation analysis proved that there was direct correlation between the the levels of HMGB1 and The number and maturation changes of dendritic cells or The number of dendritic cells surface TOLL-4 receptor in tumor, spleens (P<0.001); between the maturation changes of dendritic cells and The number of dendritic cells surface TOLL-4 receptor(P<0.001).Conclusion:Cryoablation could destroy tumor cells to release a large number of HMGB1,the HMGB1 could stimulate the body to produce a great quantity HMGB1, although gradually lowered by time. HMGB1 could activation of dendritic cells by activative TOLL-4 receptor, Cryoablation could induce tumor-specific immune response.
