objective:To observe the effect of Rho-ROCKII inhibitor c functional recovery after spinal cord injury in rats. Methods:(1)140 healthy adult female SD rats were randomly divided into 4 groups(35/group), among the total, group A,B,C were complete paraplegic which located in T8-T10 made by SD method and insert a soft catheter in cavitas subarachnoidealis of L4 planar. GroupD only openned the vertebral plate and didn't injure spinal cord. GroupA was treated by Y-27632, groupB was control group treated by PBS, groupC was control group treated by nothing, group D was Sham group. (2)Y-27632 and PBS were injected by microinjector in the groupA and B after lh of injury(12μL,14d). (3) HE and TUNEL dyeed in 8,24,168 h to observe the changing of tissue and calculate apoptosis cells with IPP 13.0 computer image analysis system; to calculate the area of growth associated protein-43(GAP-43)with IPP 13.0 computer image analysis system in 1d,1,2 w; to detect latency and wave amplitude of SEP in 3 and 8 w after SCI.;to inject fluorescent tracer through stereotaxis instrument in 6 w and observe axonal growth in 8 w after SCI; to make BBB scale in the 3,6,8 w after SCI. (4) All of the experimental results were demonstrated by x±s and calculated by SPSS 13.0 statistics software with the method of variance analysis. Results:(1)HE staining:compared to groupB,C, population of neurocyte and colloid in groupA was larger, degenerating cell and RBC was smaller, edema and cavitation was littler. (2) TUNEL staining:the mean counted apoptosis cells in group A,B,C,D at 8h was 50.5±4.70,58.9±2.46,58.5±2.1,8.3±1.44; 24h, 45.0±4.00,55.4±4.94,54.5±3.11,8.3±1.44; 168h,9.9±1.58,15.7±1.50, 15.6±1.68,8.3±1.44。the counted of group A were significant less compared to groupB,C,D, of group D were significant less compared to groupB,C, between groupB and group C were not significant different (3) GAP-43:the mean area in each group at 1d was 76.9±3.71, 43.3±3.99,42.3±4.77,37.3±2.69; lw,104.9±3.83,70.1±3.57,68.2±3.34, 37.3±2.69; 2w,159.1±4.73,94.5±3.91,96.13±3.56,37.27±2.69. the area of group A were significant less compared to groupB,C,D, of group D were significant less compared to groupB,C, between groupB and group C were not significant different. (4) SEP, the latency:3w, 30.2±0.57,36.0±1.41,35.9±0.89,14.5±0.79; 8w,23.1±1.52,28.5±1.27, 27.9±1.43,14.5±0.79; wave amplitude:3w,3.58±0.084,14.18±1.604, 1.64±0.089,1.32±0.192; 8w,6.80±0.274,3.00±0.339,2.80±0.374, 14.18±1.604. both latency and wave amplitude of group A were significant less compared to groupB,C,D, of group D were significant less compared to groupB,C, between groupB and group C were not significant different each time. (5) Fluorescent tracing:the mean fluorescent area in each group was74.8±22.49,35.0±1.58,33.7±1.30, group A were significant less compared to groupB,C, groupB and group C were not significant different. (6) BBB scale:the mean scale in each group at 3w was 8.4±0.99,5.0±0.85,5.2±0.86; 6w,9.8±1.15,5.6±1.18, 6.1±0.74; 8w,12.6±1.40,7.9±1.06,7.9±0.96. group A were significant higher than group B,C, groupB and group C were not significant different. Conclusion:Y-27632 could decrease secondary injury of CNS, improve the regeneration of injured axon, it's profit to the recovery of functional recovery after SCI.
