Objective To establish the CXC chemokine ligand 16 (CXCL16) transgenic mouse model, and to study the role of CXCL16 on lipid variability in vivo.Methods CXCL16 expression plasmid cxcl16-trans containing Flag fragment was successfully reconstructed, and was micro-injected into fertilized zygotes from C57BL/6J×CBA mice to prepare transgenic mice. Progeny were screened for the presence of the transgene by PCR. 10-week-old F3,F4 and F5 generation of CXCL16 transgenic mice(n=4) and wild-type mice(n=4) were used in the study. The protein expression level of CXCL16 was identified by Western Blot, and the lipid of blood serum was assessed by ultra-violet ray spectrometer.Results 1. A total 26 mice were found to be CXCL16 positive from the 171 founder generation by PCR and 31 CXCL16 transgenic mice were obtained from 71 F5 generation mice.2. Compared with wild-type mice, the CXCL16 transgenic mice showed an increased cholesterol (TG),triglyceride (TC) ,low density lipoprotein cholesterol(LDL-C), and a decreased high density lipoprotein cholesterol (HDL-C) (P<0.05).Conclusions1. CXCL16 transgenic mice contain Flag fragment have been successfully developed.2.. The CXCL16 transgenic mice showed an increased cholesterol (TG),triglyceride (TC),low density lipoprotein cholestero(lLDL-C), and a decreased high density lipoprotein cholesterol (HDL-C) (P<0.05).
