Objective: To investigate the effect of the mutual regulation of Pokemon and E2F-4 in HepG2 cell, and to find the new gene target for the tumor therapy.Methods: Constructed E2F-4 expression plasmid and different fragments of E2F-4 promoter, the Pokemon expressing plasmid and E2F-4 promoter,E2F-4 expressing plasmid and Pokemon promoter were transfected into HepG2 cell by the method of lipos- ome transfection, using dual luciferase reporter gene assay detected E2F-4,Pokemon transcriptional activity ; real time PCR assay detected E2F-4 mRNA after transfected Pokemon expressing plasmid; ChiP was used to detect the binding ability of Pokemon with the core sequence of the E2F-4 promoterResults: The Pokemon expressing plasmid repressed the activety of the different fragments of E2F-4 promoter , and the E2F-4 expressing plasmid repressed the activety of the different fragments of the Pokemon promoter. After transfected the Pokemon expressing plasmid, the expression of E2F-4 mRNA lever was inhibited . Pokemon represented a direct binding ability with the promoter of E2F-4.Conclusions: Pokemon expressed in HepG2 cell, repressed the transcription of the E2F-4,represented a direct binding ability with the promoter of E2F-4, maybe reduced the mRNA level of E2F-4, inhibition the expression of the E2F-4.E2F-4 repressed the transcription of proto-oncogene Pokemon in HepG2 cell.
