| Objective:To study the expression and significance of integrin-linked kinase(ILK) in the renal tubular epithelial to mesenchymal transition induced by albumin, and the relationship between ILK and transforming growth factor-β(TGF-β).Methods:Human kidney proximal tubular epithelial cell line (HK-2) was used in this study. HK-2 cells were divided into blank control group(DMEM),anti-TGF-β1 neutralizing antibody control group(DMEM+ anti-TGF-β1 neutralizing antibody 5ug/ml), albumin-induce group (DMEM+ albumin 5mg/ml)and TGF-β1-inhibited group(DMEM+ albumin 5mg/ml+ anti-TGF-β1 neutralizing antibody 5ug/ml). Each group cells were collected at 0, 12, 24 and 48 hours. Morphological changes of the HK-2 cells were observed by an inverted phase contrast microscope. The expressions of ILK mRNA, TGF-βl mRNA, FN mRNA andα-SMA mRNA were detected by reverse transcription polymerase chain reaction(RT-PCR).Results:Albumin(5mg/ml) induced ILK, TGF-βl, FN andα-SMA mRNA expression in HK-2 cells gradually increased from 0h to 48h and significantly higher than the blank control group(p<0.01). When the role of TGF-βl was inhibited by anti-TGF-βl neutralizing antibody, albumin induced-TEMT was suppressed and the expression of ILK was decreased(p<0.01). Pearson correlation analysis showed that ILK mRNA expression had positive correlation with TGF-βl, FN andα-SMA mRNA Pearson correlation analysis showed that ILK mRNA expression had positive correlation with TGF-βl,FN,α-SMA mRNA (r=0.944, r=0.989, r=0.974, p<0.01).. Conclusions:Albumin can up-regulated the expression of TGF-βl, ILK,α-SMA, FN mRNA in HK-2 cells. The levels of TGF-βl, ILK,α-SMA, FN mRNA were increased in a time-dependent manner within 48 hours. Anti-TGF-β1 neutralizing antibody might partly inhibit all of those changes induced by albumin. ILK is a TGF-βl critical downstream mediator for phenotypic transformation of renal tubular epithelial cells into mesanchymal cells induced by albumin.
