Objeetive:Detection of the EGFR (epidermal growth factor receptor) expression and EGFR mutation situation in ovarian cancer. Characteristics of its expression and mutation studies.To study the relationship between the mutation and expression of EGFR and carcinogenesis of ovarian cancer.To address the targeting of EGFR in ovarian cancer provide a theoretical foundation.Methods: 1. Paraffin-bedded tissues were obtained randomly from 55 patients who underwent surgery in the Second Affiliated Hospital of Nanchang University Hospital and Tumor Hospital of Jiangxi Province.There were 12 cases of normal ovary tissue, 12 cases of benign ovarian tumors and 31 cases of malignant ovariant tumor.The immuno-histochemical method were used.To detect the expression of EGFR. Statistical analysis was performed using x2 test and Fisher exact test. The value of P less than 0.05 were considered significant and analysis of clinical data.2. Paraffin-bedded tissues were obtained randomly from 60 ovarian cancer patients. Using PCR amplification and gene sequencing to detect EGFR tyrosine kinase coding regions of 19,21 exon in ovarian cancer, the results were analyzed using x2 test and analysis of clinical data.Results:1. EGFR expression in ovarian cancer was 89.7%,significantly higher than 50% of benign ovarian tumors and normal ovarian tissue of 0% (P<0.05).2. EGFR in ovarian serous adenocarcinoma positive expression rate was 88.9%,in ovarian non-serous adenocarcinoma positive expression rate was 84.6%.the difference was not statistically significant.From differentiation ,the expression rate of EGFR is 62.5% in highly differentiated and moderately differentiated, 95.7% in low differentiated (x2 =5.805,p<0.05)). From TNM sort (I-IV stage), the expression rate of EGFR is 73.3% in I-II stage,100% in III-IV stage (x2 =4.899,P<0.05).3. The gene mutations was not detected in 19,21 exon of EGFR in ovarian cancer.Conclusions:1,The more latter in clinical stage was and the cell differentiation was, the higher positive rate of EGFR was. The positive rate of EGFR was correlated with histodifferentiation and clinical stages of ovarian cancer, but was uncorrelated with the pathological type.2,Ovarian cancer is not detected EGFR gene mutations in 19 and 21 exon. The positive rate of EGFR was not correlated with the gene mutations in 19 and 21 exon of EGFR.
