Abstract/Summary:
Objective:To study the effective mechanism of QufengZhijingFang on transcri ption factor T-bet,GATA-3 and TNF-αin asthmatic rats. Methods:Sixty Wister rats were randomly divided into asthmatic model group,normal control group and treatment groups(hypsi-dosage,mid-dosage and low-dosage),they were given medicine respectiveiy. We weighted the T-bet,GATA-3expression by RT-PCR,weighted the TNF-α,level by Enzyme-linked- Immuo-Sorbent Assay (ELISA) . Results: The GATA-3 expressed in model group higher than normal control group and treatment groups,it has statistical significance (p<0.05);low-dosage group expressed higher than hypsi-dosage group,it has statistical significance (p<0.05);hypsi-dosage group compared with normal control group,there is no statistical significance (p > 0.05);mid- dosage compared with low-dosage,there is no statistical significance (p>0.05);mid- dosage and low-dosage group higher than normal control group,it has statistical significance (p<0.05);mid- dosage group compared with hypsi-dosage group,there is no statistical significance(p>0.05).The T-bet expressed in model group sig- nificantly higher than normal control group and treatment groups,it has statistical significance (p<0.05);low-dosage group lower than hypsi-dosage group,it has statistical significance (p<0.05);mid-dosage and hypsi–dosage group higher than model group,it has statistical significance (p < 0.05).The TNF-αexpressed in blood serum,model group higher than normal control group,it has statistical significance (p<0.05);hypsi - dosage group lower than model group,it has sta- tistical significance(p<0.05).Conclusion:QufengZhijingFang can reduce GATA-3 and raise T-bet expressed in peripheral blood,so that the balance Th1/Th2 can revert,mean- while,it can reduce TNF-αexpression.So that it can achieve the aim of treat asthma in asthmatic rats.
