ObjectiveTo explore the effects of of quercetin on ovarian follicular development and ovarian life span in rats.Method(1) Effects on fetal and neonatal rat ovaries :the experiment was divided into three groups.①Intragastric perfusion group, pregnant rats were treated with quercetin (40mg/kg.d) by intragastric administration on day 10.5 after pregnancy till delivery.②Intraperitoneal injection group,female neonatal rats were intraperitoneally injected with quercetin (40 mg/kg.d) within 12 hours after birth;③Control group ,those female newborn rats without any treatment. Female neonaltal rats were killed at postnatal day (PD) 1, PD2, PD4, ovarian development were observed.(2)The effect in early aged rats: the 12-month-old rats were administered with gastrogavage in a dose of 40mg/kg quercetin for 4 consecutive months (the control group was fed with normal saline of same volume instead), ovarian morphology were observed. In addition,vaginal smears were used to observe estrous cycles patterns of all early aged rats,uterus and ovaries were weighed to calculate their coefficients.Results(1) The percentage of unassembled follicles decreased and primordial follicles increased in the 1-day-old rat's ovaries of intragastric administration group.(2) In the 2-day-old rats , Compared with the control group,the percentage of unassembled follicles decreased and development follicles increased in intragastric administration group , the percentage of unassembled follicles also decreased but primordial follicles increased in intraperitoneal injection group.(3) The number of antral follicles increased in 15-month-old rats of administration group. (4) There were no significant changes in early-aged rats of administration group involved in the total number of survival follicles,the coefficients of ovarian and uterine and the patterns of estrous cycles. ConclusionQuercetin may promote ovarian follicular development and maturity, but have no effect on the ovarian life span. ObjectiveTo evaluate the influences of genistein to ovary in adult or pre-menopausal ovarian follicular development and ovarian life span.Methodthe 3-month-old rats were administered with gastrogavage in a dose of 160mg/kg genistein for 4 consecutive weeks, and the 11-month-old rats were administered by the same dose and methods for 4 consecutive months. The control groups were fed with normal saline of same volume instead. Meanwhile, vaginal smears were collected to observe all rat estrous cycle patterns, Hematoxylin–eosin (HE) staining was used for the ovarian histological observation.Results(1) in 3-month-old genistein-treated group, the percentage of primordial follicles increased, whereas, antral and atretic follicles decreased compared with control rats(p<0.05) (2) in 11-month-old genistein-treated group, the numbers of primordial follicles, secondary follicles and the total of survival follicles increased (p <0.05). (3)Vaginal cytology showed that regular estrous cycles of 11-month-old rats gradually disappeared and became prolonged or irregular with age. However, at the age of 15 months, 9 rats still had estrus(irregular cycles) in genistein-treated group, but only 8 rats in control group,estrous disappearing in the rest of the rats.Conclusiongenistein alter ovarian follicular differentian in adult and early aged rats through inhibiting primordial follicle development, decreasing follicles atresia, and increasing follicle survival, which may benefit to prolong the reproductive life of ovary.
