Objective: The purpose of these experiments is to detect genetic marker related to the human aerobic capacity. Therefore, we have studied A/G single nucleotide polymorphism of VDR gene using normal TaqMan probe. And we have analyzed the association between the SNP and the maximal aerobic capacity of elite swimmers of Han nationality from Shanghai.Methods: This research selected 90 elite swimmers from Shanghai Han nationality as the research objects and 85 normal subjects from Shanghai Han nationality as the controls. We investigated A/G polymorphism of VDR gene by using normal TaqMan probe. And then, maximal O2 consumption, relative maximal O2 consumption, maximal CO2 consumption, maximal respiratory quotient, maximal exercise ventilation, maximal breath frequency, maximal heart rate , maximal power, maximal exercise time, maximal watt exercise and maximal oxygen pulse were investigated by a maximal test with graded increase and a test with breath by breath. Finally, we have performed a relative analysis between the distribution of A/G polymorphism of VDR gene and maximal athletes' aerobic capacity. Results:(1) We find that the frequency of AG genotype is higher than the frequency of AA and GG genotype in both normal subjects and elite swimmers from Shanghai Han nationality. The distribution of A/G polymorphism of VDR gene is no significant difference in different gender of two groups. The distribution of A/G polymorphism of VDR gene have a significant difference between normal subjects from Shanghai Han nationality and Han nationality normal people from north China, China Kazak, China Tibetan and China Uigur. Compared to Japanese athletes, elite swimmers' genotype and allele distribution from Shanghai Han nationality are remarkably different. In addition, there is no obvious difference in genotype and allele distribution between the elite swimmers and normal subjects from Shanghai Han nationality.(2) Master swimmers are higher than first-degree swimmers in the same genotype in maximal oxygen consumption and maximal oxygen pulse. Between AG genotype of male master swimmers and first-degree swimmers, this difference is significant. Maximal O2 consumption, maximal exercise ventilation, maximal oxygen pulse, maximal power and maximal exercise time in the AG genotype of master swimmers are higher than AA and GG genotype. It is suggested that AG genotype swimmers may have a better aerobic performance than others.Conclusions: The frequency distribution of VDR gene SNP has obvious difference between Shanghai Han nationality and North China Han nationality. It's suggested that the frequency distribution of VDR gene SNP may have difference in different area. The single nucleotide polymorphism of VDR gene AG genotype varies significantly among different grade swimmers. The grade higher, the distribution of AG genotype is higher. Considered the higher frequency and the better aerobic capacity in AG genotype elite swimmers, we think that AG genotype adolescent people may be first choice from the porpective of molecular genetics if we choose good swimmer in future.
