A Preliminary Study On The Literature Research And Experimental Research On The Treatment Of Ovarian Premature Senility Combined With Traditional Chinese Medicine And Western Medicine

[Objective]This study reviewed the recent-ten-years literatures of the premature ovarian failure (POF) therapy by the integrative medicine. The prescription drugs and the medicine law were summated to guide the clinical treatment and improve the efficiency of POF therapy. Moreover, in order to provide novel therapy approach for POF, the cooperation method of Erxian Recipe and stem cellular transplant therapy was employed to treat the rats with POF.[Methods]1. Literature research:By the retrospective method, the POF literatures published by CNKI between January 2006 to December 2015 were collected. The prescription drugs were summarized and the Excel database was also set up. The western medicine therapy, the etiology and pathogenesis in Traditional Chinese Medicine (TCM) and the treatment method were reviewed through the statistical analysis.2. Experimental research:This study was based on the animal models after the chemotherapy-induced POF, and the Chinese Medicine, i.e., Erxian Recipe, was cooperated with umbilical cord mesenchymal stem cells (UCMSCs). The estrous cycle variation, vital signs, distribution of stem cells in rats’ovary and the variation of serum estradiol and follicle stimulating hormone were observed. The main objective was to find a better way to cure POF.[Results]1. Literature research:It was summarized according to 52 literatures based on the source and retrieval method that (1) The dysfunction of kidney, liver and spleen was the basic pathogenesis and deficiency of qi and blood stasis was an important process. The basic method of TCM therapy for curing POF includes: ① adjustment of zang-fu function including reinforcing kidney, and invigorating spleen and soothing liver.② reinforcing kidney assisted with blood-activating and stasis-dissolving was the common therapy, which was coupled with antipyretics. (2) The alterative therapy by hormones include medroxyprogesterone acetate plus Premarin, and medroxyprogesterone acetate plus Progynova. (3) The modern integrated traditional Chinese and Western medicine for POF was different with various syndromes of POF, and the major TCMs contained four types.2. Experimental research:(1) The rats after chemotherapy-induced POF behaved weight loss, coat color darking, disorder and disappear estrous cycle, serum estradiol decrease and follicle stimulating hormone increase. (2) The distribution of UMSCs in rats’ovary after 15 and 30 days transplantation can be found under fluorescence microscope. UMSCs were mostly distributed in ovarian cortex and medulla ovary for 15 and 30 days, respectively. Generally, stem cells had a uniform distribution. (3) The comparison of serum hormone after 15 days treatment indicated that the E2 value of three therapy groups increased but no statistical difference was observed in UMSCs treated group (p>0.05). The TCMs and cooperative group showed statistical increase (p<0.05) and the cooperative group indicated most significant increase. The FSH decrease showed statistical difference for all the three groups (p<0.05). Statistical difference was observed for E2 value after 30 days treatment (p<0.05, p<0.01) and the cooperative treatment increased most significantly. All the three treatment groups showed FSH decrease with statistical difference (p<0.05), and the TCM and cooperative groups showed most significantly difference (p<0.01).[Conclusion]1. Literature research:The dysfunction of kidney, liver and spleen was the basic pathogenesis for POF. In the treatment, Western and Chinese medicine focused on hormone replacement and reinforcing the kidney to strengthen the spleen, respectively. Method of integrated traditional Chinese and Western medicine was much better.2. Experimental research:Method coupled with Erxian Recipe and UMSCs performed much better on the improvement of serum estradiol value.
