Study On Substance Basis Of Damp - Heat Syndrome "Different Disease Syndrome" Based On Metabonomics

Purpose:To reveal the biological basis in chronic hepatitis B viral hepatitis(CHBV), non-alcoholic fatty liver disease(NAFLED) and chronic glomerulonephritis(CGN) with same sydrome of "dampness-Heat Internal Smoldering(SR) " Zheng from the perspective of metabonomics.Method:The patients with three different diseases including CHBV with SR Zheng(YS), NAFLED with SR Zheng(FS) or CGN with SR Zheng(SS) were collected, and CHBV without Zheng (YZ) and healthy controls (N) were seted as control groups, each group 30 cases. The fasting serum samples of YS, FS, SS, YZ and N were detected by UPLC-QTOFMS and GC-TOFMS, then, the results were analyzed by the statistical analysis methods, such as Variance Analysis, Principal Component Analysis and Partial Least Suares Regression. Based on those, we are eager to find out the generality and specificity of metabolic material in three different disease with SR.Results:Through the comparisons of different diseases with SR Zheng, we could find out different information from different diseases. So remove disease information of three kinds of diseases, we found out SR Zheng related materials, there were 27 kinds of substances in YS,28 in FS and 24 in SS; Compared the different substances in three diseases with SR Zheng, we observed the change of Inosine, Uridine, Aspartic Acid, Aspartic Acid, Oleic Acid glyceride and Lactate together with the three diseases of SR Zheng. Furthermore, the YS, FS, SS, YZ and N could be distinguished, each two groups differed in metabolism; in those groups, there was a high similarity betwween YS and YZ. And FS and N also had a high similarity, but SS was different from other groups; To make further comparision of each two groups, compared with the normal group, we could find 58 kinds of different substances in YS such as Beta-Alanine,55 kinds of different materials in FS, such as Cystine and 48 kinds in SS, such as Ethanolamine; In those three different diseases with SR Zheng, there were 63 kinds of different substances betwwen YS and FS,57 betwwen YS and SS and 57 betwwen FS and SS.Conclusion:Based on metabonomics, in three different diseases with SR Zheng, not only find out their respective materials, but also detect out common substances related to SR Zheng. Thus, those results could provide proof for the theory "Different dseases with the same syndrome have common substaces in the level of metabonomics".
