Part I:Establishment and evaluation for wear particle-induced rats femur periprosthetic osteolysis modelObjective Establish wear particle-induced rats femur periprosthetic osteolysis model to simulate the pathological progress of particle-induced prosthetic aseptic loosening, using Micro-computed tomography to evaluate the degree of rats periprosthetic osteolysis.Methods 30 adult SD rats were randomly divided into 3 groups(10 per group):a blank group, a sham group, and a model group. Polyethylene particles and titanium rods were implanted into the right femur of rats in model group; the sham group accepted surgery but no polyethylene particles and titanium rods; the blank group did not undergo any process. After 12 weeks, the right femur of the animals were dissected and scanned using micro-computed tomography (micro-CT) to acquire three-dimensional imagery of their cancellous bone micro structure and for analysis.Results Micro-CT three-dimensional imagery showed that in model group the bone micro structure of rats femur were damaged badly with the bone trabecula being sparser thinner and less connected when compared to blank group and sham group. Meanwhile, model group exhibited a significant increase in the BMD、BV/TV、Tb.N and Tb.Th, when compared with the others. while the SMI、BS/BV、Tb.Sp and TB.Pf were significantly lower. There was no significant difference between blank group and sham group.Conclusion Wear particle-induced rats femur periprosthetic osteolysis model is simple, repeatable and could more accurately mimic the pathological process of prosthesis aseptic loosening.Part Ⅱ:Assessment of the impact of Youguiyin Decoction on wearparticles-induced periprosthetic osteolysis using Micro-CT scanningObjective To investigate the treatment inhibiting effect of Youguiyin Decoction on wear particles-induced periprosthetic osteolysis, using Micro-computed tomography for three-dimensional evaluation, and to provide the theoretical basis for the prevention and treatment of artificial joint loosening by Youguiyin Decoction.Methods 40 adult SD rats were randomly divided into 4 groups(10 per group):a model control group, a normal saline group, a Youguiyin Decoction group, and a Alendronate sodium group. Polyethylene particles and titanium rods were implanted into the right femur of rats in each group. Three days after surgery, rats in normal saline group were administered 2ml 0.9% normal saline per 100g (according to rats’weight) a day; in Alendronate sodium group, rats were treated by 2ml Alendronate sodium(suspension of distilled water and tablets) with a dose of 0.1mg/(kg·d) once daily; in Youguiyin Decoction group, rats were administered 2ml volume drugs per 100g through intragastric administration, three times a week. After 12 weeks, the right femur of the animals were dissected and scanned using micro-computed tomography (micro-CT) to acquire three-dimensional imagery of their cancellous bone micro structure and for analysis.Results Micro-CT three-dimensional imagery showed that in Youguiyin Decoction group and Alendronate sodium group the bone micro structure of rats femur were significantly improved with the bone trabecula being denser, thicker and more connected when compared to model control group. Meanwhile, they also exhibited a significant increase in the BMD、BV/TV、Tb.N and Tb.Th. but a decrease in the SMI、BS/BV Tb.Sp and TB.Pf. There was no significant difference between model control and normal saline group.Conclusion Youguiyin Decoction can effectively inhibited periprosthetic osteolysis induced by wear particle rats femur and improve bone microstructure.
