Nitrite and nitrate can be widely found in the soil, water and plants in nature. It is inevitable to dietary intake of nitrite and nitrate and excessive intake of them will produce serious harm to health. The study of nitrite and nitrate mainly focused on vegetables and food, and the research about the content of nitrite in traditional Chinese medicine rarely reportedThis dissertation consists of two parts. The first part is the literature review:the main source of nitrite and nitrate in plants, limited standard and analysis method were summarized. The second part is the experimental section. The ion chromatography of traditional Chinese medicine for the assaying of nitrite, nitrate and for the assaying six kinds of ion chrinmatography was established, and through analysis and Compare of content of difference for scutellaria, salvia miltiorrhiza and other 11 kinds of Chinese medicinal materials of nitrite and nitrate in the first place.Meanwhile six kinds of ion chromatography for the determination of negative ion content in the blood of rats was established,in the process of analysis and comparison of aging rats anion content changes and the intervention effect of dwarf lilyturf, the main results were as follows: 1. Analysis method and content comparative of nitrite and nitrate in common Chinese traditional medicineTo establish a determination method for nitrite and nitrate in salvia miltiorrhiza, radix scutellariae and other 11 kinds of medicinal herbs by ion chromatography method. Extracted by ultrasonic, Purified by C18 column, directly sampling after centrifugal filtration.The method with a average recovery between 94.5%-104.8% was proved to be rapid, sensitive and accurate.Here we clearly demonstrate that:there is a significant difference in nitrate and nitrate content in root, stem and leaf salvia miltiorrhiza of different varieties; nitrate content in different organs of Salvia miltiorrhiza plant:stem> root> leaf; there is a significant difference in nitrate and nitrate content of salvia miltiorrhiza in different origin; Continuous cropping obstacles and fertilizer can affect the accumulation of nitrite and nitrate in salvia medicinal materials, bigger influence on the nitrate.Nitrite and nitrate in the existence of proprietary Chinese medicine containing salvia miltiorrhiza, and content in different manufacturer production of salvia miltiorrhiza proprietary Chinese medicine has obvious difference; there is a significant difference in nitrate and nitrate content of radix scutellariae, patchouli, honeysuckle and other medicinal materials in different origin; Most of the nitrite and nitrate of Chinese herbal medicine would all dissolve into the water decoction after 20-min decoction.2. Analysis method and content comparative of anionic in common herbal medicinesTo establish chromatographic method on common herbal medicines, including Patchouli, Chinese wolfberry, honeysuckle, Chuanxiong, Yuan Hu, turmeric, saffron, Zeeland, Achyranthes, for determining ion content, including Cl-、NO2、NO3-、PO43-、SO 42-、Oxalate ions.The method is ultrasonic assisted extraction. It is purified then by C18 SPE column, and then directly into the sample after centrifugation, finally with external standard. Different medicines choose a different eluent concentration in order to achieve the six kinds of anion chromatography peaks well separated. The results showed that linear relationship, the precision and stability of Chinese common herbal medicines in ions are very good, and the sample recovery rate was 93.8%-105.6%, meeting the test requirements. Anion assay results indicated that the difference of six anion contents is very significant in different parts of medicinal herbs and in different origin. Cluster analysis showed that it can be used anion content differences to distinguish the different origin of Chinese herbal medicines.The determination results about the content of anion in chinese herbal medicines were used commonly showed that the content of chloride ion in different medicinal parts of chinese herbal medicines has significant difference, the content of grass herbs was the most, at 8544.4 mg/kg, the content of root-stock medicinal material is the lowest, at 2356.8 mg/kg, the order of content was grass > flower> root> rhizome class; the content of phosphate in different medicinal parts of chinese herbal medicines has significant difference, the content of flower herbs was the first,at 3656.0 mg/kg, the content of root-stock medicinal material was the lowest, at 1251.1 mg/kg, the order of content was flower> grass> root; the content of sulfate in different medicinal parts of chinese herbal medicines has significant difference, the content of flower herbs was the first, the content of root-stock medicinal material was the lowest, the order of content was root-stock> grass> root; the content of Oxalic acid salt in different medicinal parts of, chinese herbal medicines has significant difference, the content of flower herbs was the most,the content of root-stock medicinal material was the lowest, the order of content was root> flower>grass> root-stock;The content of Cl-, NO 2,NO3-, PO43-, SO42-, oxalate ions in different origin samples from nine chinese medicinal materials have significant difference;The Anion in chinese traditional medicine from different regions can be for clustering analysis.3. content analysis method of anionic in rats blood based on aging model and the intervent function of radixTo establish chromatographic method on content determination of anionic in rats blood. The precision、accuracy and stability about this method is good, was suitable for analyzing the content of inorganic anion in rats blood. The determination results about the content of anionic in rats blood showed that the content of six kinds of anion content has a significant difference in different treatments, dwarf lilyturf intervention brought rats blood group and the dwarf lilyturf compound by nitrite,the content of nitrite and nitrate decreased in rats blood significantly because of the intervention effect of ophiopogon japonicas group...
