Objective:With the development of global industrialization, the air pollution gets increasingly serious. Sulfur dioxide (SO2) is one of the most common pollutants in the atmosphere. China’s SO2 emissions reached 23 million 700 thousand tons in 1995. This huge number transcended Europe and the United States. The harm of SO2 pollution on human health have already drawn experts’attention in the early last century. Epidemiological studies have shown that SO2 pollution can increase the risk of the hospitalization rate and mortality of the respirator cardiovascular and cerebrovascular system diseases. In recent years, many scholars studied on the toxicological effect of SO2. Through establishing animal model of SO2 inhalation, they found that oxidative damage of SO2 on tissue is an important part of the toxicological effect.SO2 can reduce the content of antioxidant, decrease the activity of antioxidant enzyme, and increase the content of lipid peroxidation. The research of SO2 should be gradually changed. Based on toxicological studies on SO2, we should start to explore the protection and treatment of SO2 toxicity. What kind of drugs can prevent or treat the oxidative injury of SO2 should be the focus of follow-up researches. We study on SO2 pollution by the theory of traditional Chinese medicine. It is different from the six evils or pestilence of the traditional Chinese medicine theory. It is a new type external toxin, it cannot be controlled and the living beings all it in. Preventing and treating it is difficult. SO2 first goes into the lungs through the nose and throat and effects on the lung’s function. Long-term toxic effects wound make the circulation of qi abd blood disorder. Then the qi stagnation, phlegm, blood stasis and other pathological changes wound emerge. We choose the respiratory system as the research object. We want to prove that traditional Chinese medicine Fei Bi Tang 2 can prevent or cure the lung injury caused by sulfur dioxide. We want to find the theory evidence of the prevention and treatment for health hazard caused by sulfur dioxide, and provide the basis for the research and development of traditional Chinese medicine for preventing and curing lung injury.Methods:We made mice exposed to SO2 (28±2mg/m3), and then took the mice of the TCM treatment group intragastric administration with Fei Bi Tang 2 for 14 days and took the mice of the TCM prevention group intragastric administration with Fei Bi Tang 2 for 28 days. Then we tested the content of propylene glycol (MDA) and glutathione (GSH), and tested the activity of glutathione peroxidase (GSH PX), superoxide dismutase (SOD) and catalase (CAT) of lung tissue. And we observed the pathological section of lung tissue.Results:1. General conditions and body weight changes:compared with the blank control group, the mice of the model group, TCM treatment group and TCM prevention group was significantly worse and their weight growth rate was significantly slowed down. Compared with the model group, the mucosal irritation symptoms of the TCM treatment group and the TCM prevention group were mild.2. The appearance of lung tissue:The lung tissues of the blank control group were pale pink, gas and had no consolidation region and the surface is smooth. The lung tissues of the model group, the TCM treatment group and the TCM prevention group were murky grey, and have some punctiform and reddish brown areas of consolidation, and the surface is not smooth.3.Histological observation:The morphology of the lung tissue of the blank control group was intact and there was no inflammatory cell infiltration in the blank control group.The alveolar wall of lung tissue in mice of the model group was widened, and the alveolar walls were damaged. There were pulmonary bullae and infiltration of inflammatory cells in the lung tissues. The alveolar walls of TCM treatment group mice got widened and were damaged as well. And there were also pulmonary bullae and infiltration of inflammatory cells in the lung tissues. but compared with model group. But compared with the model group, the degree is lighter. The alveolar wall broadening of TCM prevention group was not obvious and the alveolar walls were damaged. There were pulmonary bulla, and a small amount of inflammatory cell infiltration in alveolar walls.4.Biochemical index detection:Compared with the blank control group, the content of MDA in lung tissue of the model group was significantly increased (P< 0.001), GSH content and CAT activity was significantly reduced (P< 0.001), the activity of GSH-Px, SOD was reduced (P< 0.05).Compared with the model group, the MDA content of the TCM treatment group was decreased, the GSH content was increased, and the activity of GSH-Px, SOD, and CAT was recovered, but it was not statistically significant.In addition, compared with the TCM treatment group, MDA content of the TCM prevention group was further reduced, GSH content and the activity of GSH-Px, SOD, CAT was further increased, but also did not have statistical differences.Conclusions:1.Fei Bi Tang 2 can promote the absorption of inflammatory exudation in the lung tissues caused by SO2 exposure.2.Fei Bi Tang 2 has a protective effect on oxidative damage of lung tissues caused by SO2 exposure.3.It may take a longer time for TCM to repair oxidative damage and restore the antioxidant system activity.
