Purpose:Observe the essence on the kidney Yang deficiency model rats induced by adenine method of pharmacodynamics index, support cellular tight junction protein JAMS and ZOS protein and tight junction dynamics control factor TGF- beta 3, Ca2 +- atpase, PKA m RNA expression level of influence.Material and method : Choose 48 healthy SPF male Wistar rats, 2 months of age, body weight 180 + 20 g, were randomly divided into control group, model group, compound Xuan Ju capsule group, Yi Jing Fang group, each group of 12. Except the control group, both groups with adenine 1 m L/(100 g · d) dose lavage for 12 consecutive days; Starting from the13 th day, the control group and model group with the amount of normal saline lavage,compound Xuan Ju and beneficial essence group with compound Xuan Ju capsule aqueous solution, and the yi jing fang one to fill the stomach, once a day, 20 days in a row.Experimental rats after anesthesia, aseptic conditions take 10 ml of blood in the abdominal aorta, gather the testis and epididymis, Using the computer sperm count rate of sperm density and sperm activity analysis system; By the methods of enzyme-linked immunosorbent(ELISA) for each group of rats serum levels of testosterone(T) for testing; Using immunohistochemical technique to JAMS protein in rat testicular tissue, ZOS protein expression; By RT-PCR technology determination of TGF- beta 3, Ca2+-atpase, PKA m RNA expression.Results:1. Compared with the control group, model group animal sperm activity rate, density and serum testosterone T were significantly decreased(P < 0.01); Compared with model group,compound Xuan Ju capsule group and yi jing fang group of sperm activity rate, density and serum testosterone T have increased significantly(P < 0.01).2. Compared with the control group, model group JAMS and ZOS protein levels significantly decreased(P < 0.01); Compared with the model group, yi jing fang group of JAMS and ZOS protein levels higher(P < 0.05), compound Xuan Ju capsule group is not obvious(P >0.05).3. Compared with the control group, model group of TGF- beta 3 m RNA expression increased(P < 0.01); Compared with the model group, yi jing fang group of TGF-beta 3m RNA expression decreased(P < 0.05), compound Xuan Ju capsule group decline is not obvious(P > 0.05).4. Compared with the control group, model group of Ca2+- atpase the expression of m RNA and PKA m RNA were lower(P < 0.01). Compared with model group, yi jing fang group of Ca2+-atpase m RNA, PKAm RNA expression increased(P < 0.05), the compound Xuan Ju capsule group increase is not obvious(P > 0.05).Conclusion:1. Adenine can reduce sperm density and vitality, and the level of serum testosterone T;While making JAMS, ZOS protein expression decreased, increasing TGF- beta 3 m RNA expression and reduced the expression of Ca2+-ATPm RNA and PKAm RNA.2. Ying Jing Fang can improve the kidney Yang deficiency model rats induced by adenine method of sperm density, sperm activity rate and the concentration of serum testosterone T,make JAMS and ZOS protein expression level increased; The high expression of TGF- beta 3m RNA has obvious inhibitory effect, the Ca2+-atpase lower expression of m RNA,PKAm RNA has obvious antagonistic effect.
