Objective:To determine the median lethal concentration (LC50) of cisplatin,5-Fu,cisplatin and 5-Fu mixed solutions in Caenorhabditis elegans (C. elegans) by the acute toxicity test, which provides reference for further sub-acute toxicity experiments.Methods:K-medium solution was used to prepare different concentration of cisplatin,5-Fu,cisplatin and 5-Fu mixed solutions as the experimental group, K-medium solution as the control group. The death and survival numbers of C. elegans were calculated after its had been exposed to the cisplatin,5-Fu, or mixed solutions 24h. At the same time, the growth and development, reproduction and movement of C.elegans was observed. LC50 and 95% CI were derived by Probits analysis.Results:① LC50 ofC. elagans was 617mg·L-1 (95% C7:601~631 mg·L-1) when it exposed to cisplatin for 24h.②The LC50 was 19600 mg·L-1 (95% CI: 18510~20717mg·L-1) in 5-Fu for 24h.③ When C. elagans exposed to mixed solutions for 24h, the LC50 of cisplatin was 589 mg·L-1 (95% CI:536~721 mg·L-1) and the LC50 of 5-Fu was 18620 mg·L-1 (95% CI:16954 22795mg·L-1).We also found that the growth and development, reproduction and movement of the C. elegans were inhibited in the experiments. With the concentration of drugs increasing, the restrain was more obviously.Conclusion:The LC50 of 5-Fu is 31.8 times higher than cisplatin, which indicated that the lethal toxicity of cisplatin is more greater than 5-Fu in C. elegans, and C. elegans is suitable for the preliminary assessment of acute toxicity experiment.Objective:To investigate the effects of body length in Caenorhabditis elegans (C. elegans) exposed to lower concentrations under LC50 of cisplatin,5-Fu, cisplatin and 5-Fu mixed solutions.Methods:According to the LC50 in part one, which were diluted from 1/10 to 1/1000 with K-medium solution,then obtained different concentration of cisplatin,5-Fu,cisplatin and 5-Fu mixed solutions as the experimental group, K-medium solution as the control group. The body length of C. elegans was observed after it had been exposed to solutions for 60h. Each concentration was random selected 30 nematode to measure the body length. The experiment was repeated three times, a total of 90 nematode (n= 90),Then the body length of C. elegans was calculated with mean and standard deviation.Results:①After 60h exposed to 0.62 mg·L-1,6.2 mg·L-1,62 mg·L-1 cisplatin solutions and K-medium solution, the body length of C. elegans in 0.62 mg·L-1 and 6.2 mg·L-1 solutions were no difference with control group, it were [(0.696±0.032) vs (0.693±0.016)/(0.691±0.020), n=90, P> 0.05]. The body length of C. elegans in 62 mg·L-1 solution was inhibited significantly, it was [(0.696±0.032) vs (0.489±0.028), P<0.01].②After 60h exposed to 19.6,196,1960 mg·L-1 5-Fu, compared with the control group, the body length of C. elegans in19.6,196,1960mg·L-1 exposed group was inhibited significantly, it were [(0.696±0.032) vs (0.511±0.012)/ (0.186±0.015)/(0.180±0.006), n=90,P<0.01].③After 60h exposed to cisplatin and 5-Fu mixed solution, compared with the control group, the body length of C. elegans exposed to (0.62+19.6) mg·L-1(6.2+196) mg·L-1, (62+1960) mg-L-1 solutions were inhibited significantly, it were [(0.696±0.032)vs(0.511±0.012)/(0.186±0.015)/ (0.180±0.006),n=90, P<0.01].④After 60h exposed to cisplatin, cisplatin and 5-Fu mixed solution, 0.62 and (0.62+19.6) mg·L-1,6.2 and (6.2+196) mg·L-1,62 and (62 +1960) mg·L-1, the body lengths of C. elegans in these groups were significantly difference by pairwise comparisons, it were [(0.693±0.016) vs (0.511±0.012), (0.691±0.020) vs (0.186±0.015), (0.489±0.028) vs (0.180±0.006), n=90, P<0.01].⑤After 60h exposed to 5-Fu, cisplatin and 5-Fu mixed solution,19.6 and (0.62+19.6) mg·L-1,196 and (6.2+196)mg·L-1,1960 and (62+1960) mg·L-1, the body lengths of C. elegans in these groups were no difference by pairwise comparisons, it were [(0.525±0.015) vs (0.511±0.012), (0.186±0.011) vs (0.186±0.015), (0.179±0.005)vs (0.180±0.006), n=90, P> 0.05].⑥After 60h exposed to cisplatin and 5-Fu solution,0.62 and 19.6m·L-1,6.2 and 196mg·L-1,62 and 1960 mg·L-1, the body lengths of C. elegans in these groups were significantly difference by pairwise comparisons, it were [(0.693±0.016) vs (0.525±0.015), (0.691±0.020) vs (0.186±0.011), (0.489±0.028) vs (0.179±0.005), n=90, P<0.01].Conclusion:Cisplatin and 5-Fu both have a developmental toxicity on C. elegans, and 5-Fu is more significantly impact on body length.
