Radix Polygoni Mulfiflori is the tuberous root of Polygonum multiflorum Thunb., a type of Polygonaceae perennial herbaceous plant that can been found in forests, hill-side or bushes. And the whole of the plant can be used as medicines, named Radix Po-lygoni Mulfiflori and Caulis Polygoni Mulfiflori. After being steamed with soya-bean milk, Radix Polygoni Mulfiflori can be prepared to Radix Polygoni Mulfiflori Praepa-rata Cum Succo Glycines Sotae. Radix Polygoni Mulfiflori can be used for regulating bowel movement and eliminating carbuncles,while the main pharmacological func-tions of the prepared root include nourishing the blood, benefiting the liver and kid-neys, strengthening the bones and muscles, blackening the beard and hair,etc. In re-cent years, many scholars at home and abroad have studied on Radix Polygoni Mulfiflori for different types of chemical constituents, such as stilbene glycosides, anthraquinones,etc.,and its pharmacological activities including anti-aging,anti-tumor, anti-atherosclerotic, neuroprotective effect and liver protection. However, the major researches have not yet systematically studied on the neuroprotective effect.The objective of this research was to study the compositions that may have neuroprotective effect in Radix Polygoni Mulfiflori. Using various separation methods such as silica gel column chromatography, ODS reversed phase chromatographyh, Sephadex LH-20 chromatographyh,preparative thin layer chromatography and preparative liquid chromatography, etc., twenty-two compounds were separated, and were confirmed to be 2,3,5,4’-tetrahydroxystilbene-2-O-(2"-O-p-hydroxybenzoyl)-β-D-glucoside(1*),2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside(2),2,4,6,4’-tetrahydr-oxy-stilbene-2-O-β-D-glucoside(3), quercetin(4), rutin(5), apiin(6), kaempferol(7), emodin(8), physcion(9), emodin-1,6-dimethyl ether(10), aloe-emodin(11),2-acetyl emodin(12), chrysophanol(13), rhein(14), co-citreorosein(15), emodin-8-O-β-D-gluco-pyranoside(16), emodin methyl ether-8-O-β-D-glucoside(17), questin(18), gallic acid(19), p-hydroxy benzaldehyde(20),β-sitosterol(21) and daucosterol(22) using 1H-NMR,13C-NMR and MS spectroscopy.2,3,5,4’-tetrahydroxystilbene-2-O-(2"-O-p-hydroxybenzoyl)-β-D-glucoside(1*) was a new compound.Aloe vera L. belongs to the Liliaceae family, and is a well-known traditional Chinese medicine that has been be used for many years. Researchers have found that the main chemical constituents are chromones and anthraquinones, and the pharmaco- logical activities includes antisepsis, antiviral, anti-tumor, immunoregulation, etc. β-secretase (BACE) is an important target relating to Alzheimer’s disease (AD), and BACE inhibitors may become medicines for AD.The previous work in our group has found that the crude extract from Aloe vera L. exhibits BACE inhibitory activity, and aloeresin D is the effective constituent. Meanwhile, extract with the contant of aloeresin D occupies 50%-90%, exhibits potent inhibitory activity as well, and they used macroporous resin column and ODS reversed phase chromatography to deal with the ethyl acetate extract from Aloe vera L., to prepare the fraction rich in aloeresin D (70%) as well as chromone. We determine the contents of aloeresin D and total chromones and anthraquinones in the procedure of the preparation, by using HPLC-DAD and external standard method, to explain the changes of the contents.
