Dyspepsia is the medical term used to describe the sense of "difficult digestion". The term refers to a very heterogenous group of symptoms, located in people’s upper abdomen, which can be divided into 2 main categories of organic and functional dyspepsia (FD). Spepsia symptom is common in the general population and can significantly affect the quality of patients’life, in which the most patients have functional dyspepsia (FD). Dyspepsia refers to a collection of complex upper gastrointestinal symptoms that is believed to be common world wide. The prevalence of functional dyspepsia is high that the symptoms impair quality of life of patients and the management of the condition remains a considerable burden on health care resources, it has been estimated that FD accounts for billions of dollars per year in direct and indirect costs in Europe and American. So physicians pay more attention on FD nowadays. As defined by this committee, an individual must have one or more of the following symptoms and no evidence of structural disease that is likely to explain the symptoms:bothersome postprandial fullness, early satiety, epigastric pain, or epigastric burning.First of all, use 2,4,5-trimethoxy benzoic acid react with thionyl chloride then we will get 2,4,5-trimethoxybenzoyl chloride. The crude product react with 2-aminothiazol-4-carboxylic acid ethyl ester to give 2-[N-(2,4,5-trimethoxy-benzoyl) amino]-4-(ethoxycarbonyl)-1,3-thiazole, DMAP as catalyst. The crude product was recrystallized from 95% ethanol, then will get refined products:2-[N-(2,4,5-trimethoxy-benzoyl) amino]-4-(ethoxycarbonyl)-1, 3-thiazole.2-[N-(2-hydroxy-3,4-dimethoxybenzoyl) amino]-4-(ethoxycarbonyl)-1,3-thiazole was synthesized from the refined products by demethylation with pyridine hydrochloride. After purification with acetic acid, react with excess N, N-two isopropyl ethylenediamine to give 2-[N-(4,5-dimethoxy-2-hydroxybenzoyl)amino]-4-[(2-diisopropyl-aminoethyl)aminocarbonyl]-1,3-thiazole.2-[N-(4,5-dimethoxy-2-hydroxybenzoyl) amino]-4-[(2-diisopropyl-aminoethyl) aminocarbonyl]-1,3-thiazole and then acylation with N, N-diisopropylethane-1,2-diamine, salt formation and recrystalization with an overall yield of 36.6%.In this paper, a new HPLC method is established to quantitatively analyze industrial acotiamide hydrochloride trihydrate and it’s intermediates based on its amphoteric structure. The separation is accomplished in a C18 column and the UV detection wavelength is 210 nm. This method contains no ion paring agent, benefiting the rapid balance of chromatographic condition and improving the analysis repeatability and maneuverability.The method can be performed for rapid detection of acotiamide hydrochloride trihydrate and used in the quality control of raw medicine.
