Objective To study the effect and significance of perfluorocarbon by intraperitoneal injections on nitric oxide(NO),endothelial nitric oxide synthase(e NOS), endothelin-1(ET- 1) in serum and aorta tissue and the pathological changes of aorta of rats. To discuss the possible protective effect and mechanism of perfluorocarbon on endothelia function of rats with atherosclerosis. To provide a part of theoretical basis for the PFC’s clinical application in the cardiovascular disease.Method The 40 male Wistar rats were randomly divided into 4 groups: control group, atherosclerosis model group, low dose of PFC group, high dose of PFC group(10rats/each group).The rats in the control group were fed with normal forage.Others were fed with high fat diet. establish the experimental rat model with AS.While modeling,the treatment groups were given different doses of PFC. Until the model was established for 12 weeks.Serum and aorta tissue were collected for being measured, and the pathological changes of aorta were observed by HE stain.Result Compared with control group, the level of NO, e NOS in model group were lower and ET-1 were higher remarkably(P<0.05). Compared with model group, the content of NO, e NOS in high dose of PFC were increased(P<0.05) and ET-1 were decreased(P<0.05);while the content of e NOS, NO in serum low dose group increased and ET-1 decreased but no obvious difference was observed. Compared with model group,ET-1 in aorta tissue decreased(P<0.05).Compared with low dose group of PFC, the content of NO, e NOS in high dose of PFC were increased(P<0.05) and ET-1 were decreased(P<0.05). Compared with low dose group of PFC, the high do se of PFC inhibited the pathological changes of aorta obviously.Conclusions Certain dose of PFC maybe protect endothelial function by increasing the synthesis of e NOS,NO, decreasing ET-1,and keeping its balance.
