Objective To investigate the synergistic neuroprtective effect of salidroside combined with mild hypothermia rats with cerebral ischemia-reperfusion injury and the mechanism.Mnethods The 10 minutes global cerebral ischemia rat model was created by the modified Pulsinelli’s 4VO method. The 200 male Wistar rats were randomly divided into five groups, including sham group(Sham), model group(Model), salidroside group(Sal), mild hypothermia group(Mild), salidroside combined with mild hypothermia group(Sal+Mild). The rats in the Sham group were performed as the IR model without the vertebral arteries cauterized or the carotid arteries occluded.IR was performed without any other handling measure in the Model group. IR was performed with the precondition of salidroside in the Sal group. The rats were intraperitoneally injected with 10mg/kg salidroside at 10:00 am each day lasting seven days, andthe IR models were made at 30 minutes after the last injection. The rats were put in the environment of 4℃ to reduce the anus temperature to 32℃ lasting 2h after the cerebral ischemia models made in the Mild group.IR was performed with the precondition of salidroside and the reducing of temperature as the Sal group and the Mild group in the Sal+Mild group. The neurobehavioral evaluation was assessed according to the NDS, moisture capacity was calculated the ratio of dry weight to wet weight, the modality was displayed by HE staining, the apoptotic cells were displayed by TUNEL staining, the SOD activity, the content of MDA in hippocampus CA1 were assessed at 24 h after reperfusion and the expression of Bcl-2 and Bax was assessed by Western blot.Results Compared with the Model group, the Neuro Deficit Scores,the brain moisture capacity, the morphological changes in the hippocampus CA1 region, the content of MDA and the expression of Bax were reduced and the SOD activity and the expression of Bcl-2 was increased in the Sal group, Mild group, Sal+Mild group(P<0.05), moreover the effect was more obvious in the Sal+Mild group(P<0.05).Conclusion Salidroside and mild hypothermia have a synergistic effect on cerebral ischemia reperfusion injury, which may be related to anti-free –radical effects and the upregulation of Bcl-2 and the downregulation of Bax.
