In order to study the genome structures and evolutional characters of mitochondrialgenome in Gomphoceridae reconstruct the phylogenetic relationship of Gomphoceridae.Inthis study,the complete sequences of Orinhippus tibetanus genome was determined usingPCR and clone methods. The structure, nucleotides conposition, genes arrangement,synonymous codons usage bias etc, of the Orinhippus tibetanus genome was analyzed indetail. Additionally,using NJ, MP, ML and Bayes methods,the phylogenetic relationship of4species downloaded from GenBank was reconstructed by concatenated13PCGs.Theconclusions were showed as follows:1.The complete mitochondrial genomes of Orinhippus tibetanus is15611bp.All genomecontents consist of the13protein-coding genes,2rRNA genes,22tRNA genes,and anA+T-rich region localized between rrnS and trnI.The trnD and trnK genes have changedpositions relative to the putative ancestral arthopod mtDNA gene arrangement.2.The base disparity in the Orinhippus tibetanus mitochondrial genomes are seriously,asthe contents of AT is74.2%.In the protein-coding genes,the AT scope is73.1%.The ATcontents of rRNA and tRNA are76.9%and72.9%,respectively.The AT content of A+T-richregion is highest,which is84.8%.3.The most used initiation codon is ATG in the Orinhippus tibetanus protein-codinggenes,then is ATT and ATC,while ATA is the least;the protein-coding genes take TAA astermination codons commonly,then is TAG and TAT.4.Using NJ,MP,ML and Bayes methods,the phylogenetic relationship of Orinhippustibetanus was reconstructed by concatenated13PCGs.The study shows the monophyly ofGomphoceridae has been well supported.The Aeropus licenti and Aeropus tibetanus are sistergroup with Gomphocerus rufus together constitute the Gomphocerinae;Pacris xizangensis asPacrinae and Orinhippus tibetanus as Orinhippinae constitute the top of the molecular tree asthe most evolutionary taxa. The results support the view of plateau locusts adaptiveevolutionary presented by Yin in1982.
