Studies on secondary metabolites of marine microorganisms have becomehotspots that international scientists pay much attention to in recent years. Marinemicroorganisms could produce various kinds of structurally novel and bioactivecompounds taking into consideration the complicated living environments and theirpeculiar metabolisms. Rich in resources and having a great capacity for producingbioactive metabolites, mangrove-derived fungi has become the most systematicallyresearched marine fungi in present.In order to find the novel bioactive compounds, this thesis used the biologicaland chemical methodology to isolate and screen the endophytic fungi from themangrove plant Bruguiear gymnarrhiza and study the secondary metabolites of2endophytic fungi. This work includes: the isolation and bioactivity test of mangroveendophytic fungi, the fermentation of selected fungi, the structural elucidation of theirmetabolites and the bioactivity tests of these secondary metabolites.13endophytic fungi were isolated from the mangrove plant Bruguieargymnarrhiza, which was collected from Dongzhai Gang mangrove reserve, Zhanjinag,Guangdong province. According to the chemical and biological tests, the66-2#strainwas discovered to possess good bioactivities and chemical diversity. And it isidentified as Penicillium sclerotiorum by the molecular biology method.On the basis of several chromatography techniques and spectral methods, the twospecies of fungi Penicillium sclerotiorum and AGR-6#strains were studied on theirsecondary metabolites in this thesis.17compounds compounds including(+)-cyclopenol,3-(dimethylaminomethyl)-1-(1,1-dimethyl-2-propenyl)indole,verrucosidin, deoxyverrucosidin, anacine, aurantiomide C,(22E)-Ergosta-5,7,22-trien-3beta-ol, ergosterol peroxide, viridicatol, viridicatin,3-O-Methylviridicatin,3R,4R-3,4,8-trihydroxy-1-tetralon, cyclo-(Tyr-Pro),4-Hydroxybenzaldehyde,3-phenylacrylamide, flavasperone, rubrofusarin B, in which(+)-cyclopenol was found as a novel compound and the cystal structure of3R,4R-3,4,8-trihydroxy-1-tetralon was first reported.At last, the bioactivities of these metabolites are tested including the anti-tumortests, the neuro-protective activity test and the anti-diabetic test. The results suggests that the compound deoxyverrucosidin possess the anti-diabetic activity.
