In this research, in accordance with the principle of polyphasic taxonomy, a systematic study had been conducted on the identification of five Bacillus-like (HB12575, HB12576, HB12037, DB13031and DB13260) by using classical taxonomy, chemotaxonomy, molecular classify, were isolated from tropical rain forest soil Jianfengling/Jiaxi and marine sediment from the northern part of Bamen Bay and Qinglan tidal inlet in Hainan.Strain HB12575:Cells are Gram-positive, aerobic, motile rods, irregular long rods. After24h growth on TSB agar, surface colonies are round, pancake, The colour of the strains clone is creamy, the edge is smooth; Optimum temperature for growth is30℃(range5-45℃) and the optimum pH for growth is7.0-9.5(range pH7.5). Growth occurs in the range of0-14%(w/v) NaCl (optimum,0-2%);The major polar lipid profiles are PIM, PI, LPG, PME and DPG. Major cellular fatty acids are iso-C14:0(4.9%), iso-C15:0(10.4%), anteiso-C15:0(51.9%), iso-C16:0(10.8%), and anteiso-C17:0(10.1%). The DNA G+C content is45.3mol%. The phylogenetic tree revealed that its16S rRNA gene had highest homology (97.6%) with Bacillus decolorationis LMG19507T, which located on the same branch. Their DNA-DNA hybridization value was91.5%. Strain HB12575was identified as Bacillus decolorationis.Strain HB12576T:Cells are Gram-positive, aerobic, motile rods, irregular long rods with a size range of0.7-1.0×2.8-3.5μm. After24h growth on TSB agar, surface colonies are pancake, edge villous, grey white smooth; Growth occurs in the range of0-15%NaCl (optimum,1-4%,w/v). Optimum temperature for growth is32-37℃(range20-55℃) and the optimum pH for growth is9.0-10.5(range pH6.0-11.0). The major polar lipid profiles are PC, DPG, PME and PE. Major cellular fatty acids are iso-C14:0(21.7%), C15:0(11.7%), ant-C15:0(33.1%), iso-C16:0(8.6%), and C12:1ω9c (10.9%). The DNA G+C content is51.7mol%. The phylogenetic tree revealed that its16S rRNA gene had highest homology (95.2%) with Bacillus okuhidensis GTC854T. Strain HB12576T represents a novel species of the genus Bacillus, for which the name Bacillus dongjiaoensis. sp. nov. is proposedStrain HB12037T:Cells are Gram-positive, aerobic, motile rods, irregular long rods with a size range of0.8-1.2×3.5-4.5μm. After24h growth on TSB agar, surface colonies are pancake, edge villous, grey white smooth; Catalase is positive. Oxidase is negative. Optimum temperature for growth is32-37℃(range20-45℃) and the optimum pH for growth is7.5-9.0(range pH5.5-11.5). Growth occurs in the range of0-15%(w/v) NaCl (optimum,1-4%). The major polar lipid profiles are PE, PME and DPG. Major cellular fatty acids are anteiso-C15:0(43.1%), iso-C15:0(5.7%), iso-C16:0(10.1%), C17:0(11.6%), and cis-C16;1(4.3%). The DNA G+C content is53.9mol%. The phylogenetic tree revealed that its16S rRNA gene had highest homology (95.9%) with Paenibacillus xylanilyticus XIL14T. Strain HB12037T represents a novel species of the genus Paenibacillus, for which the name Paenibacillus bamenwangensis sp. nov. is proposed.Strain DB13031T:Cells are Gram-negetive, aerobic, motile rods, irregular long rods with a size range of0.5-0.7×2.0-3.0μm. After24h growth on TSB agar, Colonies are round, colonies raised, The colour of the strains clone is creamy, the surface is smooth; Optimum temperature for growth is30-37℃(range10-53℃) and the optimum pH for growth is7.5-9.5(range pH5.5-11.5). Growth occurs in the range of0-4%(w/v) NaCl (optimum,0-2%);The major polar lipid profiles are PME, LPG, PI, PC, PG and DPG. Major cellular fatty acids are iso-C14:0(3.6%), iso-C15:0(4.2%), anteiso-C15:0(54.8%), iso-C16:0(12.1%) and C16:0(13.7%). The DNA G+C content is49.7mol%. The phylogenetic tree revealed that its16S rRNA gene had highest homology (98.3%) with Paenibacillus barcinonensis BP-23T, which located on the same branch. Their DNA-DNA hybridization value was38.1%. Strain DB13031T represents a novel species of the genus Paenibacillus, for which the name Paenibacillus jiaxiensis sp. nov. is proposed.Strain DB13260T:Cells are Gram-negetive, aerobic, motile rods, irregular long rods with a size range of1.0-1.6×3.0-5.0μm. After24h growth on TSB agar, Colonies are round, The colour of the strains clone is creamy, the surface is smooth; Optimum temperature for growth is30-37℃(range5-40℃) and the optimum pH for growth is8.5-9.0(range pH6.0-10.5). Growth occurs in the range of0-4%(w/v) NaCl (optimum,0%); The major polar lipid profiles are PME, PI, LPG, PE, PIM, PC and DPG. Major cellular fatty acids are iso-C14:0(7.1%), C14:0(3.0%), iso-C15:0(5.9%), anteiso-C15;0(43.9%), iso-C16:0(19.6%), and C16:0(3.5%). The DNA G+C content is53.7mol%. The phylogenetic tree revealed that its16S rRNA gene had highest homology (98.3%) with Paenibacillus anaericanus MH21T, which located on the same branch. Their DNA-DNA hybridization value was60.4%. Strain DB13260T represents a novel species of the genus Paenibacillus, for which the name Paenibacillus jianfenglingensis sp. nov. is proposed.
