Shrimp is one of the most important food allergens. Tropomyosin is itsmajor allergen. Wherein Pen a1, contains five antibody binding regions, has beenidentified as the only major shrimp allergen. It has high sequence identities andsimilarities compared with most of the shellfish species. Shrimp allergic reaction mainlybelongs to the Type I allergy which is caused by IgE. At present, studies on polyclonaland monoclonal IgE with response to Shrimp Allergen have been reported. However, noone involved in preparation of small molecule IgE to the same allergen. This articleattempts carry out research both on obtaining cell materials and gene cloning. It set up asimple and effective immune system of shrimp allergen in vitro and identified five pairsof primers of κand λchain. The obtained gene fragments contain complete FRs,CDRs functional structure, and the identity can be more than90%compared with thedatabase. In addition, this paper also demonstrates the importance of immunization invitro to the cloning of antibody variable regions. The study will provide strong technicalsupport to the further building of shrimp allergens scFv and the shrimp allergenantibody library. As well as the research on antibody gene rearrangement rule from themolecular level.
