Opal gene is encoded in cell nuclear, and the protein it encodes belongs to mitochondrial shape proteins. It plays an important role in mitochondrial morphology, respiration and apoptosis. Besides, it is related with the synthesis of cytotoxic steroid in vivo and the inhibition of cytotoxicity of cancer drugs, which influences many diseases.The aim of the study was to investigate OPA1mRNA expression in mouse uterus during early pregnancy, delayed implantation and activation, pseudopregnancy, artificial decidualization and hormone regulation model by in site hybridzation methods. OPA1mRNA sigal was almost undetected on day1of pregnancy. The sigal was expressed in luminal epitheliuma and glandular epitheliuma of uterus in hormone regulation model, and the intensity of sigal was reduced in estrogen treatment and increased in progesterone treatment on2h,8h and24h of treatment. From day2to4of pregnancy, the sigal was only expressed in luminal epitheliuma, and intensity was higher on day2and4. On day5of pregnancy, the sigal was expressed in luminal epitheliuma and glandular epitheliuma and stromal cells surrounding the implantation site of uterus. In the activation of implantation model, the situation of expression was similar to the situation of day5of pregnancy. However, in the delayed implantation model, the signal was barely detected in uterus. From day1to5of pseudopregnancy, the sigal was both expressed in luminal epitheliuma and glandular epitheliuma of uterus, but its intensity was almost unchanged. On day6and7of pregnancy, the sigal was mainly detected in decidua area, whose intensity didn’t change. On day8of pregnancy, the sigal was expressed throughout the uterine matrix, and the intensity enhanced. In the uterine tissue of artificial decidualization, the sigal was strongly expressed in primary decidual area. While in the control group, the signal only appeared in luminal epithelium and glandular epithelium with low intensity.Based on the results, the expression of OPA1mRNA is specific during early pregnancy. The sigal detected in luminal epithelium and glandular epithelium may be controlled by hormone regulation. While the expression change in stromal cells may be affected by embryo implantation and decidualization.
