Effects Of The Recombinant Eukaryotic Expression Vector PcDNA3.1(+)-ChIL-4to The Immunological Function In Chickens

There is large scale poultry farming in China. The poultry disease prevention plays an important role in the process of production. At present, the vaccines are usually used to prevent disease. In order to play vaccine effectiveness better, the research of immune adjuvant become one of the hot spot. Cytokines adjuvants have become the focus of the researchers due to its specific functions and strong biological activity and low side effect. This study explore the immune adjuvant function of IL-4, carried out the following test:design primers and cloning gene of chicken IL-4according to the gene sequence of chicken IL-4which is in the NCBI (login AJ621249). ChIL-4genes is cloned successfully after enzyme digestion, the PCR identification and sequencing, confirmed that the homology of100%with chicken IL-4gene sequence alignment in NCBI. Recombinant eukaryotic expression vector pcDNA3.1(+)-ChIL-4was successfully constructed finally. Animal testing pointed6group:normal saline control group, the vaccine control group, pcDNA3.1(+) empty vector control group, and3different doses of pcDNA3.1(+)-CWIL-4experimental group, respectively,50μg,100μg,200μg. Discusses the affected of IL-4to chickens activity of peripheral blood T/B lymphocytes percentage, ND, IB antibody levels induced by the vaccine, the immune organs (spleen) index. Test results show that adding pcDNA3.1(+)-ChIL-4experimental group can improve chicks activity of T/B lymphocytes levels in the blood, can improve the ND, IB, antibody levels induced by the vaccines significantly, raise chicken spleen index in a certain stage, and200μg dose group effect is relatively obvious. These results showed that the recombinant eukaryotic expression vector pcDNA3.1(+)-ChIL-4can improve the immune function of chickens, enhance the immune effect of vaccine and it can be used as immune adjuvant.
