The plants of Macleaya are widely distributed in China, which contends plentifulalkaloids. It plays an important role in agriculture. Studies showed that the alkaloidsin the plants played certain effects on antitumor activity and anti-AIDS in recent years.So it is important to set up a rapid and efficient method for extraction and separationof the alkaloids from the Macleaya in large scale production.The study uses methods of ultrasonic wave extraction and inner ebullitionfor extraction and comparison of the main constituent-alkaloids from Macleayamicrocarpa (Maxim.) Fedde, setting up a method-Using high performance liquid chromatography to determine the contents of sanguinarine and chelerythrineindicating the total alkaloid content, and uses AB-8resin to separate the extract from Macleaya microcarpa. The results were presented as the followings:1The ultrasonic process for extracting alkaloids from Macleaya microcarpa withMethanol was studied. Comparison of the content of alkaloids in different parts of rawmaterial, experiment showed that the extraction of the leaves was the highest, than thestems,at least in the roots.2To establish a HPLC method for the determination of sanguinarine and chelerythrine content analysis method. HPLC analysis was carried out on Extend-C18column and the acctonitrile-0.3%acetic acid solution (23:77) as the mobilephase, the flow rate at1.0mL/min, the column temperature was25℃and thedetection wavelength at277nm. According to the standard curve detection resultsof drawing two alkaloids standard. Sanguinarine regression equation is:Y=84950x-681.81(R2=0.9994)chelerythrine regression equation is:Y=101506x-49.332(R2=0.9994).The measurement results are reliable.3Decompressing inner ebullition was chosed to extract alkaloids from Macleayamicrocarpa. By examining the various extraction conditions the optimum extractionprocess is determined: the concentration of desorption solvent ethanol80%for30min,th extraction concentration was50%ethanol, the extraction temperature was50°C, theextraction pressure was-0.07MPa, the extraction time was8min. Using this conditiontechnology, the extraction rate of sanguinarine was0.74mg/g and the extraction rate ofchelerythrine was1.38mg/g. Through the comparison of the decompressing inner ebullition and ultrasonic extraction results showed that, the decompressing innerebullition method was better than the ultrasonic extraction.4Different concentrations of ethanol elution ingredient Macleaya microcarpa were isolated by AB-8resin and to detect the isolates using infrared spectroscopy. Therefore, using different ethanol concentrations can be separation the alkaloids from Macleaya microcarpa.
