| Due to continuously increasing rates of cancer and increasing cell pathologicalchanges in recent years, cells research has very important influences. In current cellseparation, there are a lot of ways, such as flow cytometry instrument, crosschromatographic separator.ect, but there are disadvantages in all those methods. Based onthat, the aim of this project is to develop a kind of instrument can quickly separate cells,which is based on the principle of laser guide cells to realize the separation of cells in highflux, high efficiency and without damage.The topic of this subject is the research of the guiding role of laser in micro-fluidiccells, which is based on the laser particle, according to the principle of momentum of a cellseparation method. Specifically it is based on the particle interaction when the light and themomentum interacting, which cause cells the motion state of change, to realize apredetermined target cell separation.In this article, it is first introduced that the related topics of background significanceand present situation at home and abroad, and the purpose of the subject, mainly illustratesthe principle of laser guide cells and force analysis; Second, COMSOL simulation softwareis used to conduct simulation experiments in various experimental conditions, includinglaser factors and factors of micro-fluidic chip, and a series of data is achieved, whichprovides theoretical data basis for the experiment; at last the experimental platform is set upto conduct the laser guiding cell experiment. During the experiment, by changing theparameters of affecting the experimental results, such as laser power, laser wavelength, withthe guide of micro fluid inside the cell, it is achieved that the goal of the single channelseparation cells at low speed. Under the existing equipment in this experiment, we separatethe yeast cells with laser-guiding in micro-fluidic chip separation. The separating speed isabout30per minute, effective separation efficiency is about70%.Above all, the subject has made some achievements, but there are still many to study,there are many disadvantages:1) micro-fluidic chip should be made into multiple channelwhich can achieve the multi-channel separation at the same time;(2) the method of lasertriggering should be developed at the same time to shorten the time required for single cellseparation, thus improve the separation speed. |
