Study On Metabolites Of Three Entomogenous Fungi

Three strains of endophytic fungi as the research object in this paper. Two strains ofPestalotiopsis fungi include L567Pestalotiopsis virgatula and Pestalotiopsis zonata392.Another one strain of Entomogenous fungus which is ACCC37776Isaria farinosa. Thesefungi have been cultivated, fermentation, extraction separation and identification theirspectrum structure. Some compounds have been made relevant bioactivity tests. L567Pestalotiopsis virgatula cultivated by fluid nutrient medium. The other two strains of fungicultivated by solid culture medium. Twenty-five compounds (including three new ones) wereisolated from three strains of endophytic fungi (L567Pestalotiopsis virgatula andPestalotiopsis zonata392and ACCC37776Isaria farinosa), using various chromatographicmethods (preparative thin-layer chromatography, silica gel column chromatography elution,reversed phase HPLC and recrystallization). Structures of these compounds were elucidatedby four spectroscopic (NMR, MS, IR and UV) analysis.Four compounds were isolated from this fungus of L567Pestalotiopsis virgatulaCompound1:2-Cyclohexene-1-propanoic acid,2,6,6-trimethyl-4-oxo-, methyl ester.Compound2*: methyl3-methyl-5-(2,6,6-trimethyl-4-oxocyclohex-2-en-1-yl).Compound3: Dimethyl ortho-phthalate. Compound4: Diethylamine. Compound2*is a newcompound. Thirteen compounds were isolated from the fungus of Pestalotiopsis zonata392.Compound5:4-hydroxy-4-methyl-2h-pyran4hydrogen-2-ketone. Compound6:7-hydroxy-5-methoxy-4,6-dimethyl-7-O-a-L-rhamnose phthalide. Compound7:Hydroxybenzoic acid. Compound8: Hydroxyphenylacetic acid. Compound9: Hydroxy-phenethyl alcohol ethanol. Compound10:3,6,8-3hydroxy-1,2,3,4-four hydrogennaphthalene1-ketone. Compound11: phenylethyl phenylacetate. Compound12:2H-Pyran-2-one, tetrahydro-5-hydroxy-6-methyl. Compound13: phydroxy benzaldehyde.Compound14*:(4-Hydroxy-phenyl)-acetic acid2-hydroxy-1-methyl-propyl ester.Compound15: succinic acid. Compound16:4-hydroxyphenethyl3-hydroxypropanoate.Compound17:4-hydroxyphenethyl acetate. Compound14*is a new compound.Eight compounds were isolated from the fungus of ACCC37776Isaria farinosa.Compound18:(3b,5a,6b,22E)-6-methoxyergosta-7,22-diene-3,5-dio. Compound19: (22E,24R)-Ergosta-4,6,8(14),22-tetraen-3-one. Compound20: Named Arinosa(1).Compound21: Named Arinosa(2). Compound22:6,8-dihydroxy-3-((1E,3E)-penta-1,3-dien-1-yl) Isochroman-1-one. Compound23:12-oxooctadecanoic acid. Compound24*:N-[(2,3-dihydroxyphenyl) carbonyl] glycine methyl ester. Compound25:2,3-dihy-droxybenzoyl-N-glycine. Compound24*is a new compound.Six compounds2*,14*,21,22,24*and25have been made high throμghput screeningmodel tests. Include (protein tyrosine esterase) CDC25B antitumor model; Dehydrogenase(hypoxanthine nucleotides) IMPDH1and IMPDH2immune inhibitor model; Protein tyrosinephosphatase SHP2inhibitor model; Protein tyrosine phosphatase PTPN11antitumor model;Dipeptide base peptidase4(DPP4inhibitor) anti diabetes model. The results showed thatCompound25and22have activity to CDC25B antitumor model. The IC50value ofcompound25and22is23.6μg/mL and19.8μg/mL respectively. Compound22againstenzymes: IMPDH1and IMPDH2, The IC50value is35.34μg/mL and11.38μg/mLrespectively. Another compounds have not showed activities. In addition, compound21hasbeen tested activity on cancer Hela cells. The results showed that compound21inhibitoryeffect on the growth of Hela, IC50=27.0μg/mL.
